鹿茸总多肽对维A酸致骨质疏松大鼠的防治作用

摘    要: 目的观察鹿茸总多肽(TVAP)对维A酸诱导大鼠骨质疏松的影响。方法通过ig维A酸70 mg·kg-1·d-1,连续14d,建立大鼠骨质疏松模型。造模同时sc TVAP 60,40,20 mg·kg-1·d-1,治疗30 d后,处死动物。取血分离血清,测定血清钙(Ca)、磷(P)、碱性磷酸酶(ALP);取左侧股骨,计算骨重系数和测定骨Ca,P含量;取右侧股骨,测定中段和下1/3处骨密度;取左侧胫骨,做骨组织形态计量测定。结果与模型组比较,TVAP明显改善维A酸诱导大鼠的骨Ca含量降低(P<0.01),骨重系数下降(P<0.001)和骨密度降低(P<0.05,P<0.01)以及骨病理形态发生的明显改变:平均骨小梁宽度(MTPT)变窄(P<0.001)、平均骨小梁间距(MTPS)变大(P<0.05)、松质骨体积(TBV)降低(P<0.01)等骨质疏松改变。但对血中反映骨代谢状况的生化指标Ca,P含量和ALP活性无明显影响。结论TVAP能纠正维A酸所致骨重建的负平衡状态,使骨量增加,骨组织显微结构趋于正常,对大鼠骨质疏松有防治作用。

Abstract:OBJECTIVE To investigate the effect of total velvet antler polypeptides(TVAP) on the osteoporotic model rats induced by retinoic acid.METHODS The male Wistar rats were given the retinoic acid 70 mg·kg-1·d-1 for 14 d intragastrically and sacrificed on day 30.At the same time,the osteoporotic rats were treated with TVAP at a dose of 60,40 and 20 mg·kg-1·d-1 subcutaneously.The effects of TVAP on osteoporosis were evaluated by serum biochemistry,bone mineral density and bone histomorphometry.The levels of Ca,P and ALP in serum were assayed.The bone weight coefficients of the rat left femurs were calculated and the contents of Ca and P in them were measured.Then the bone mineral densities of the mid and the below 1/3 of right femurs,and the bone histomorphometry of the left cibias were assayed.RESULTS Compared with the model group,the bone Ca content(P<0.01),bone mineral density(P<0.05,P<0.01) and bone weight coefficient(P<0.001) were significantly increased in TVAP group rats.After intake of TVAP,the mean trabecular plate thickness(MTPT)(P<0.001),the mean trabecular plate interspace(MTPS)(P<0.05) and the trabecular bone volume(TBV)(P<0.01) were improved obviously,compared with that of the osteoporotic model rats.There were no changes for the biochemical indexes reflecting the bone metabolism such as the contents of Ca,P and the activity of ALP in blood.CONCLUSION TVAP appears a remedial activity against negative-balance of bone remodeling induced by retinoic acid.TVAP can increase bone mass and improve the structure of cancellous bone.So TVAP has a preventive and therapeutic effect on osteoporotic rats.
