当归多糖组分的高效液相色谱分析

摘    要: 目的:分析当归多糖AP-Ⅰ,AP-Ⅱ,AP-Ⅲ,AP-Ⅳ各组分的重均相对分子质量分布及其电荷特性;探讨多糖类生物大分子的组效关系研究方法。方法:采用高效凝胶过滤色谱法、阴离子交换色谱法、二极管阵列检测器进行检测;改良苯酚-硫酸法测定糖分部;Serva Blue-G 染料结合法测定蛋白质分部。结果:当归多糖AP-Ⅰ,AP-Ⅱ,AP-Ⅲ,AP-Ⅳ分别由4~5个具有不同重均相对分子质量分布及电荷特性的多糖组分组成;4种多糖中还分别含有一定量的游离或结合蛋白质。结论:当归多糖AP-Ⅰ,AP-Ⅱ,AP-Ⅲ,AP-Ⅳ为组成成分不同的多糖;该方法可为多糖组效关系研究提供简便、实用的分析方法。

Abstract:OBJECTIVE:To analyze the distribution of weight average molecular weight (r) of four Angelica polysaccharides isolated from Angelica sinensis (Oliv.)Diels:AP Ⅰ,AP-Ⅱ,AP-Ⅲ,APⅣ and determine their characters of electric charge,in order to provide a practical analytical method for studying the constituent activity relationship of polysaccharides. METHODS:Two kinds of high performance liquid chromatography methods were employed,including high performance gel filtration chromatography for r determination and high performance anion exchange chromatography for electric charge analysis.Improved phenol sulfuric acid method combined with ELISA reader were used for the polysaccharide fractions determination,and Sever blue G dye combined method for protein fractions as well. RESULTS:Each of the four Angelica polysaccharides,AP-Ⅰ,AP-Ⅱ,AP-Ⅲ and AP-Ⅳ,was composed of 4~5 polysaccharide constituents with different r distribution and characters of electric charge;and there were small amount of different free or combined proteins in the four Angelica polysaccharides. CONCLUSION:The four Angelica polysaccharides,AP-Ⅰ,AP-Ⅱ,AP-Ⅲ and AP-Ⅳ,are different from each other in r and character of electric charge;this method is practical and reliable for the study of constituent activity relationship of polysaccharides.
