氯唑沙宗片的人体药物动力学研究

 采用反相ＨＰＬＣ法测定人血清中氯唑沙宗浓度。线性范围０．２６±６２．５２μｇ，ｍｌ￣（－１），高、中、低３种浓度回收率分别为９７．０％±２．３％，９７．１％±３．５％，９３．８％±３．０％（ｎ＝５），日内、日间ＲＳＤ均＜４．８％。以本法测定８例．怒愿受试者氯唑沙宗血药浓度，并计算分析药代动力学参数，结果单剂量口服８００ｍｇ氯唑沙宗后，药时曲线呈一室模型。Ｔ＿（ｍａｘ）＝１．６３±０．４３ｈ，Ｃ＿（ｍａｘ）＝１９．７１±３．９０μｇ·ｍｌ￣（－１），ＡＵＣ＿（０～∞）＝８２．２０±１１．３１ｍｇ·ｈ·Ｌ￣（－１），Ｋａ＝０．９２±０．３８ｈ￣（－１），Ｋｅ＝０．５４±０．１１ｈ￣（－１），Ｔ＿（１／２）＝１．３１±０．２４ｈ，Ｖｄ＝１８．６３±３．１５Ｌ，Ｃｌ＝９．９２±１．５５Ｌ·ｈ￣（－１）。

Pharmacokinetics of chlorzoxazone in human body

The serum concentration of chlorzoxazone was determined by RP-HPLC method。The linear range was 0.26～62.52 μg·ml￣(-1).The recovery rate of high,middle,low concentra-tions was 97.0%±2.3%,97.1%±3.5%,93.8%±3.0%,respectively。Coefficients of inter-dayand intra-day variations were less than 4.8%。The serum concentrations of chlorzoxazone weredetermined in 8 volunteers。 The pharmacokinetic parameters were calculated and analyzed.After asingle oral dose of chlorzoxazone 800 mg had been given to each volunteer,the pharmacokineticsof chlorzoxazone was shown by one compartement model.Tmax=1. 63±0.43h,Cmax=19. 71±3. 90μg·ml￣(-1),AUC0～∞=82.20±11.31 mg·h·L￣(-1),Ka=0.92±0.38h￣(-1),Ke=0.54±0. 11 h￣(-1),T1/2=1.31±0.24h,Vd=18.63±3.15 L,Cl=9.92±1.55L·h￣(-1)。
