枸杞多糖-1对羟自由基所致小鼠肝线粒体损伤的作用

摘    要: 目的 研究枸杞多糖(lycium barbarum polysaccharides,LBP)组分1(LBP1)清除羟基自由基(·OH)的效果及其对·OH所致小鼠肝线粒体氧化损伤的作用效果。方法 用Fe2++H2O2或Fe2++VC系统产生·OH;用水杨酸法研究LBP1清除·OH的效果;用硫代巴比妥酸法测定小鼠肝线粒体丙二醛含量、用以DPH为荧光探剂的荧光偏振法测定小鼠肝线粒体膜流动性、以及用分光光度法测定小鼠肝线粒体膨胀程度研究LBP1抗·OH氧化损伤的效果。结果 LBP1可以清除·OH,减少·OH所致丙二醛的产生,抑制·OH所致膜流动性下降和减轻·OH所致肝线粒体膨胀程度,并呈量效关系。结论 LBP1具有清除·OH和抑制·OH所致小鼠肝线粒体氧化损伤的作用。

Abstract:OBJECTIVE To study the scavenging effect of LBP1 (one component of polysaccharides from Fructus Lycii,the fruit of Lycium barbarum L.) on hydroxyl radicals (·OH),and the protective effect of LBP1 on the mice liver mitochondria injured by ·OH.METHODS ·OH were generated from Fe2++H2O2 or Fe2++VC.The scavenging effect of LBP1 on ·OH was detected by salicylic acid method.To study the protective effect of LBP1,the content of MDA was measured by TBA method.The swelling extents of mice liver mitochondria were detected by spectrophotometric method,and the membrane fluidity of mice liver mitochondria was measured by the fluorescence polarization assay with DPH fluorescence probe.RESULTS LBP1 had the ability to scavenge·OH,decrease the generation of MDA,inhibit the decrease of mitochondria fluidity induced by ·OH,and alleviate the swelling of mitochondria induced by ·OH.CONCLUSION LBP1 had the effects of scavenging ·OH and inhibiting the mice liver mitochondria injury induced by ·OH.
