丹皮酚减少大鼠局灶性脑缺血再灌注损伤后一氧化氮和内皮素的生成

摘    要: 目的观察丹皮酚对脑缺血再灌注损伤一氧化氮(NO)和内皮素(ET)生成的影响。方法采用大脑中动脉内拴线阻断法(MCAO)制备大鼠局灶性脑缺血再灌注损伤模型,测定脑缺血1h再灌注12及24h后脑组织匀浆和血清中NO以及血浆中ET水平。结果大鼠脑缺血1h再灌注12及24h,脑组织匀浆和血清中的NO、ET水平均显著升高,丹皮酚(2.5,5,10mg·kg-1·d-1,po)给药7d,可显著降低脑缺血再灌注12和24h血浆ET及脑组织匀浆诱生型NOS(iNOS)活性和NO水平。结论丹皮酚减少脑缺血再灌注损伤时NO及ET生成,可能是其保护脑缺血损伤的作用机制之一。

Abstract:OBJECTIVE To investigate the effects of paeonol on nitric oxide and endothelin production in focal brain ischemia-reperfusion injury rats. METHODS The NO and ET levels of brain homogenate,serum and plasma were measured with a model of reversible middle cerebral artery occlusion(MCAO) without craniectomy in rats. RESULTS The levels of NO and ET were increased significantly in ischemia-reperfusion rats compared with sham rats. Paeonol decreases the activity of iNOS,levels of NO in brain homogenate and decreases the plasma levels of NO and ET in 12 and 24 h following reperfusion compared with ischemia-reperfusion injury rats. CONCLUSION The results suggested that the effect of paeonol decreasing NO and ET production may play an role on amelioration of focal brain ischemia reperfusion injury.
