赤芍与白芍的化学成分含量比较研究

摘    要: 目的通过对不同产地赤芍和白芍主要化学成分的含量测定及比较,进一步完善芍药野生品与栽培品药材的质量控制标准。方法采用高效毛细管电泳法(HPCE)的胶束电动色谱(MEKC)分离模式,测定了16个产地20个赤、白芍样品中的4个有代表性的活性成分:芍药苷、苯甲酸、D-儿茶素和没食子酸,并比较各成分之间的比例关系。结果①野生芍药根(赤芍)中芍药苷含量大于6%,D-儿茶素含量高于0.05%。未加工的栽培芍药根中芍药苷含量小于6%,未检测到D-儿茶素。在同样条件下,加工后的栽培芍药根(白芍)中芍药苷含量下降了37%~56%,仍未检测到D-儿茶素,苯甲酸的变化最大,下降达83%~92%。②内蒙古多伦赤芍与大、小兴安岭所产赤芍在化学成分的种类和含量上存在差异。③不同产地、特别是道地与非道地的白芍药材之间,化学成分的比例有所不同。结论运用HPCE技术,可以快速准确地对野生与栽培芍药根的活性成分差异进行定量分析。从而为区别同为芍药来源的赤芍和白芍的临床应用提供实验依据。

Abstract:OBJECTIVE To determine the contents of paeoniflorin,D-catechin,benzoic acid,gallic acid of Paeoniae Radix collected in different areas respectively,and to improve the quality standard of Radix Paeoniae Rubra and Radix Paeoniae Alba from Paeonia lactiflora Pall.. METHODS The contents of four active constituents in 20 commercial samples of Paeoniae Radix,which were collected from 16 habitats,were successfully analysed by micellar electrokinetic chromatography (MEKC). RESULTS①MEKC chromatograms were obviously different between wild and cultivated groups of Paeoniae Radix.The contents of paeoniflorin and D catechin in wild group were more than 6% and 0.05%,respectively,while the paeoniflorin in cultivated group was less than 6.0%,and D catechin was not detected.②The constituents of Radix Paeoniae Rubra planted in the Duolun were different from that in the “Da xiao Xing'anling”.③The components of Radix Paeoniae Alba in genuine place was also different from that in non genuine place. CONCLUSION With HPCE technology,quantitative analysis of active constituents in wild and cultivated groups of Paeoniae Radix was performed accurately.
