染料木素对雌激素缺乏诱发的大鼠骨质疏松症的作用研究

摘    要: 目的观察染料木素对实验性去卵巢大鼠诱发的骨质疏松症模型的治疗作用。方法SD雌性大鼠行去卵巢术后,随机分为正常对照组,模型组,骨疏康(GSK)组,尼尔雌醇组,染料木素大、中和小剂量组,每组10只。各组动物给予盐酸四环素25mg·kg-1连续3d,同时给予治疗用药,剂量为GSK 5g·kg-1,尼尔雌醇为1.5mg·kg-1,染料木素分别为18,9,4.5mg·kg-1,以上给药除尼尔雌醇每周1次外其他各组均为每日1次。给药后12周后进行相关指标的测定。结果①染料木素4.5,9mg·kg-1连续给药12周可使其股骨干重、灰重、骨钙及股骨和腰椎骨密度显著增加,与模型组比较具有显著差异(P<0.05或P<0.01);②染料木素4.5,9mg·kg-1连续给药12周可显著改善模型大鼠的股骨及椎骨最大载荷、最大桡度、弹性载荷、弹性桡度(P<0.05或P<0.01);③染料木素低、中剂量用药12周后可明显增加模型大鼠骨小梁面积百分比(BV/TV%)、骨小梁厚度(Tb.Th)和骨小梁数目(Tb.N),减小骨小梁间隔(Tb.Sp)(P<0.05或P<0.01);④染料木素低、中剂量给药12周后,可使模型大鼠的血清ALP水平下降,尿Ca/Cr、Pyd/Cr水平降低(P<0.05或P<0.01)。结论染料木素对模型动物的骨密度、骨生物力学及相关生化指标都具有改善作用,具有较好的抗骨质疏松作用。

Abstract:OBJECTIVE To investigate the effect of genistein on osteoporosis in ovariectomized rats. METHODS The 23-week-old female rats were ovariectomized (the control group were shamed) to form the model rats, then the rats were divided into seven groups randomly as follows: control group, model group, GSK group(5.0 g·kg-1), nilestriol-treated group (1.5 mg·kg-1 twice per week), and genistein-treated groups at different dosage of high dose(18.0 mg·kg-1), medium dose (9.0 mg·kg-1) and low dose(4.5 mg·kg-1). The bone sample was collected and the parameters were determined after 12 weeks. RESULTS The thighbone weight, ash weight of the thigh, bone calcium, and the bone mass density of femur and lumbar were increased by genistein at dose of 4.5 and 9 mg·kg-1 after 12 week treatment (P<0.05 or P<0.01). The biomechanics including of maximum loading, maximum strain, flexibility loading and flexibility strain of femur were enhanced by 4.5 and 9.0 mg·kg-1 genistein (P<0.05 or P<0.01). The area percentage, thickness and numbers of bone trabecula were increased significantly and saeptum of bone trabecula was decreased in the groups treated by 4.5 and 9 mg·kg-1 genistein (P<0.05 or P<0.01). The level of ALP in serum and the level of Ca/Cr and Pyd/Cr in urine were decreased in the groups administered with genistein at dose of 4.5 and 9 mg·kg-1 (P<0.05 or P<0.01). CONCLUSION Genistein could improve the bone mass density, biomechanics and correlated biochemical indicators ovariectomized rats and could be used for osteoporosis treatment.
