重组人内抑素抑制佐剂性关节炎大鼠滑膜组织中新生血管生成及机制

摘    要: 目的观察重组人内抑素(recombinant human endostatin,rh-End)对佐剂性关节炎(adjuvant-induced arthritis,AA)大鼠滑膜组织新生血管生成的抑制作用及其部分机制研究。方法采用弗氏完全佐剂(Freund's complete adjuvant,CFA)诱导的AA大鼠模型,观察滑膜组织中新生血管生成的病理学改变;免疫组织化学法检测血管内皮细胞生长因子(vascular endothelial growth factor,VEGF)的表达。实时荧光定量PCR对VEGF的表达进行定量检测。结果rh-End能抑制AA大鼠的继发性足肿胀,对滑膜组织中过度生成的新生血管有抑制作用。Rh-End能显著减少AA大鼠关节滑膜组织中高表达的VEGF。结论重组人内抑素可显著抑制AA大鼠滑膜组织VEGF的表达及新生血管生成。

Abstract:OBJECTIVE To investigate the effect of recombinant human endostatin on the angiogenesis in adjuvant arthritis synovium tissue.METHODS Adjuvant arthritis rat model was induced by a single intradermal injection of 100 μL of CFA on the first day.The volume of rat hind paw was measured by means of volume meter.The polyarthritis score and histological change were evaluated.The expression of VEGF was measured by immunohistochemical method.VEGF mRNA was evaluated by Real-time quantitive-PCR.RESULTS rh-end attenuated the severity of arthritis on both second hind paw volume and the polyarthritis score,as well as improved the arthritic status histologically in AA rats.Simultaneously,rh-end inhibited the expression of vascular endothelial growth factor in synovial tissues.The therapeutical effect of rh-end was related to the inhibition of synovitis and angiogenesis.CONCLUSION rh-end significantly inhibited the expression of VEGF in AA rat synovium as well as angiogenesis.
