白藜芦醇抗炎作用机制的初步研究

摘    要: 目的观察白藜芦醇的抗炎作用并对其抗炎机制进行初步探讨。方法采用切除双侧肾上腺大鼠足肿胀、冰醋酸致大鼠胸膜炎及角叉菜胶致大鼠气囊炎3种炎症模型,观察白藜芦醇对不同炎症模型的抗炎作用,并测量白细胞数量、蛋白质含量、前列腺素E2(PGE2)、一氧化氮(NO)、超氧化物歧化酶(SOD)、丙二醛(MDA)、超氧阴离子(O2.-)含量变化。结果白藜芦醇高、中剂量组可减轻去双侧肾上腺大鼠足肿胀程度;减少了冰醋酸致大鼠胸膜炎渗出液量和蛋白质含量,使血清NO含量降低,SOD活性增强;抑制了角叉菜胶致大鼠气囊炎模型中白细胞游出及蛋白质渗出,降低PGE2,NO,MDA,O2.-生成量。结论白藜芦醇具有明显的抗炎作用,其抗炎作用不依赖于肾上腺的存在,可能与其降低血管通透性、抑制白细胞游出,抑制PGE2、NO等炎症介质产生及增强清除氧自由基、抗脂质过氧化能力有关。

Abstract:OBJECTIVE To study the anti-inflammatory mechanisms of resveratrol.METHODS Hind paw edema induced by egg albumin was measured in bilaterally adrenalectomized rats.The chemotaxis of leucocytes,the volume of exudate,the activity of SOD and the content of NO in serum were respectively measured in rats with acute pleuritis induced by the injection of glacial acetic acid.The number of leucocytes and the content such as total protein,PGE2,NO,MDA,and O·2 were examined in the exudate of rats with acute air-pouch synovitis induced by sc injection of 25 mg·kg-1 carrageenan.RESULTS Resveratrol has a significantly suppressive effect on hind paw edema in adrenalectomized rat induced by egg albumin.Resveratrol significantly inhibited the leucocytes and exudates infiltration,decreased the production of NO in serum,and elevated the activity of SOD in pleuritis rats serum.Furthermore,resveratrol reduced the migration of leucocytes and the exudation of protein,and inhibited the production of PGE2,NO,MDA and O·2.CONCLUSION Resveratrol has an anti-inflammatory effect,which does not depend on the existence of adrenal glands.It may be related to inhibiting the production of inflammatory mediators and free radicals.
