苦参碱对皮肤角质形成细胞IL-17R、p-p38 MAPK和炎症因子的影响

摘    要: 目的 考察苦参碱(matrine,Mat)对皮肤角质形成细胞(keratinocytes, KC)系HaCaT细胞白细胞介素-17受体(IL-17R)、p-p38 MAPK表达的影响,及其对HaCaT细胞分泌白细胞介素(IL)-6、IL-8、MIP-3α等炎症因子的影响。方法 采用RT-PCR方法检测Mat对皮肤KCIL-17R mRNA表达的影响;采用流式细胞术检测Mat对HaCaT细胞IL-17R蛋白含量的影响;采用Western blot方法检测Mat对HaCaT细胞表达p-p38 MAPK的影响;ELISA法检测Mat对HaCaT细胞分泌炎症因子IL-6、IL-8及MIP-3α的影响。结果 Mat可降低HaCaT细胞表达IL-17R和p-p38 MAPK,以及降低HaCaT细胞IL-6、IL-8、MIP-3α等炎症因子的分泌。结论 Mat可通过抑制IL-17R的表达,阻止p38 MAPK的磷酸化过程而具有抗炎作用。

Abstract:OBJECTIVE To investigate the effect of matrine on the expression of IL-17R, p-p38 MAPK in skin keratinocyte cell line HaCaT cells, and on the level of IL-6, IL-8, MIP-3α secreted by HaCaT cells. METHODS RT-PCR method was applied to analyze the expression of IL-17R mRNA in HaCaT cells treated with matrine. Flow cytometry was used to analyze the IL-17R protein content. The expression of p-p38 MAPK was determined by Western blot method. The levels of inflammatory factors IL-6, IL-8 and MIP-3α were analyzed by ELISA. RESULTS Matrine decreased the expression of IL-17R and p-p38 MAPK in HaCaT cells, and the level of IL-6, IL-8 and MIP-3α secreted by HaCaT cells. CONCLUSION Matrine can inhibit the expression of IL-17R to prevent the phosphorylation of p38 MAPK, and inhibit the expression of IL-6, IL-8, MIP-3α, and so matrine has anti-inflammatory effect.
