摘    要:目的:建立同时测定大鼠血浆中油酸和亚油酸含量的气相色谱法。方法:采用毛细管气相色谱法,色谱柱为OV-17毛细管柱(30m×0.32mm×0.50μm),柱温220℃,氢火焰离子化检测器(FID)检测。结果:该法能将油酸甲酯和亚油酸甲酯有效分离,分别在23.80μg.mL-1~2380μg.mL-1(r=0.9996)、24.51μg.mL-1~2451μg.mL-1(r=0.9997)线性关系良好,高、中、低3个浓度的样品提取回收率在75.83%~84.17%,方法回收率在95.85%~107.1%,日内和日间精密度RSD均≤4.7%。结论:本法简单、准确,可作为大鼠血浆中油酸和亚油酸成分的检测方法,为进一步研究鸦胆子油及其制剂的药代动力学奠定基础。

Abstract:Objective:To develop and validate a gas chromatography(GC) method for the simultaneous determination of oleic acid and linoleic acid in rat plasma.Methods:Chromatographic separation was performed on OV-17 capillary column(30m×0.32mm×0.50μm) with a temperature of 220℃,and a flame ionization detector.Results:The GC method had shown good chromatographic resolution for Methyl oleate and Methyl linoleate.Linearity was tested ranging from 23.80 μg·mL-1 to 2380 μg·mL-1(r=0.9996) and 24.51μg·mL-1 to 2451 μg·mL-1(r=0.9997).The extraction recoveries for two components were between 75.83% and 84.17% and the method recoveries were between 95.85% and 107.1%.The intra-and inter-day RSD were less than 4.7%.Conclusion:The proposed method is simple,accurate and suitable for the determination of oleic acid and linoleic acid in rat plasma and further pharmacokinetics study.
