逍遥散对慢性束缚应激大鼠相关脑区NMDAR2A和NMDAR2BmRNA基因表达的调节作用

摘    要:目的:观察海马及杏仁核N-甲基D-天冬氨酸(N-methyl-D-aspartic acid(NMDA)受体NMDAR2A(NR2A)和NMDAR2B(NR2B)的蛋白在束缚应激状态下的mRNA表达变化及道遥散的作用。方法:使用捆绑的方法制作束缚应激动物模型,并用逍遥散进行干预,分别于7天后和21天后检测模型大鼠海马CAI区、CA3区、齿状回(DG)、杏仁核NMDA受体NMDAR2A和NMDAR2B的蛋白mRNA表达的情况。结果:NR2AmRNA和NR2BmRNA在海马的CA1,CA3,DG和BLA都有不同程度的基因转录。NR2AmRNA在CA3,DG和BLA区,7天和21天模型组的表达均下调(P0.05和P0.01),在CA1区不明显。NR2BmRNA在CA1,CA3,DG和BLA区,21天模型组的表达均下调(P0.05和P0.01),在CA1和BLA区7天模型组表达不变。逍遥散对CA3和DG区的NR2AmRNA和NR2BmRNA有明显的调节作用,在CA3和DG区7天和21天的表达均上调(P0.05和P0.01)。而在CA1区对NR2AmRNA和NR2BmRNA没有调节作用。其中以CA3区和DG区的调节作用最明显,这里可能是逍遥散的调节靶点。结论:逍遥散在海马和杏仁核区对慢性束缚应激引起的神经传递的改变有明显的双向调节作用,可以影响突触可塑性,以适应机体的功能,主要靶点在CA3和DG区。

Abstract:Objective:To observe the change of N-methyl-D-aspartic acid(NMDA)receptor subunit and related regulatory(NMDAR2Aand NMDAR2B) protein mRNA expression in the hippocampus and amygdale in immobilization stressed rats and effect of Xiaoyaosan(XYS)on them.Methods:Immobilization model rats were established by binding for 3h per day and intervened with XYS,the expression of NMDA receptor subunit NMDAR2Aand NMDAR2B protein mRNA expression in model rats′CAI and CA3 region of hippocampus,dentate gyrus and amygdala:were detected on day7and day21 after modeling.Results:NMDA receptor subunit NMDAR2Aand NMDAR2B mRNA expression were all in CA1,CA3,DGand BLA.On day7and day21 models,NR2AmRNA expression decreased in CA1,CA3and BLA(P0.05 and P0.01),but not well in CA1.On day21 models,NR2BmRNA markedly lowered in CA1,CA3,DGand BLA(P0.05 and P0.01),and on day21 models,they were invariant in CA1and BLA.On day7and day21 Xiaoyaosan(XYS),NMDAR2Aand NMDAR2B protein mRNA expression were significantly increased in,CA3 and DG(P0.05 and P0.01).but they were constant in CA1.Result,XYS significantly regulated the expression of NMDAR2A and NMDAR2B mRNA in CA3 and DG,and modulation target spot.Conclusion:The change of cranial nerve were distinct double monitor in the hippocampus and amygdale in immobilization stressed rats and effect of Xiaoyaosan(XYS)on them,which effected nerve plasticity and fit in with the function of the organism,mostly in CA3和DG region.
