温阳活血方对单侧输尿管梗阻大鼠肾脏NF-кB和TGF-β_1表达的影响

目的:探讨核转录因子-кB(NF-кB)、转化生长因子-β1(TGF-β1)在肾间质纤维化中的表达情况及温阳活血方的干预作用。方法:采用单侧输尿管结扎(UUO)肾间质纤维化模型。将大鼠随机分为6组,即:模型组、温阳活血治疗组、桃仁治疗组、肉苁蓉治疗组、蒙诺治疗组、假手术组,假手术组只分离但不结扎输尿管。术后2周处死大鼠,检测尿β2-MG,观察肾组织病理改变,并应用免疫组织化学方法检测肾组织NF-кB和TGF-β1的表达。结果:模型组肾组织表现为纤维化病理改变,正常组肾小管上皮细胞、肾间质细胞胞浆NF-кB和TGF-β1有较弱的表达;模型组呈强阳性表达;温阳活血治疗组病理改变较模型组纤维化程度减轻,且NF-кB和TGF-β1的阳性表达较模型组显著减弱,有统计学差异(P<0.05)。结论:温阳活血方可能通过抑制NF-кB和TGF-β1的过度表达而减缓肾纤维化病程进展。

Investigation of NF-кB and TGF-β_1 Expression in Rats Kidney after Unilateral Ureteral Obstruction with Semen Persicae

Objective: To investigate the expression of nuclear factor-kB in Rats with renalinrerstitial fibrosis and the mechanism protect of Wenyang Huoxie recipe.Methods: Unilateral Ureteral Obstruction rats animal mode was used.The rats were randomly divided into 6 groups: monopril treatment group,semen persicae treatment group,Wenyang Huoxie recipe treated group,saline cistanche treated group,Sham-operated group(SOR),Sham-operated rats had their left ureters manipulated but not ligated.Renal tissue were examined by light microscopy at the 2th week after operation.Immunohistoche mistry was applied to measure the expression of nuclear factor-кB and TGF-β1 in the renal interstitium.Results: Fibrosis was observed in the renal interstitium of rats with obstructive nephropathy,the expression of NF-кB and TGF-β1 in the normal operation group is low in the renal tubular epithellal cell and renal interstitium cell.Incomparison with narmal group,the expression of NF-кB and TGF-β1 in the of mode group markedly increased(P<0.05);Incomparison with mode group,semen persicae group could significantly decrease the expression of NF-кB and TGF-β1(P<0.05);no significant difference was detected between semen Wenyang Huoxie recipe treatment group and monopril treatment group.Conclusion:Semen pesicae may suppress fibrosis development,at least in part via down regulating NF and TGF-β1 expression.
