南星骨痛凝胶贴膏对兔膝骨关节炎关节形态及血清IL-1、TNF-α、MMP-13、TIMP-1表达的影响

目的观察南星骨痛凝胶贴膏对兔膝骨关节炎关节形态及血清IL-1、TNF-α、MMP-13、TIMP-1表达的影响,探讨南星骨痛凝胶贴膏治疗兔膝骨关节炎可能的作用机制。方法 18只健康6月龄新西兰兔随机分成3组:空白组(A组)、模型组(B组)、骨痛膏治疗组(C组). BC组用3%木瓜蛋白酶关节腔内注射造模后C组给予南星骨痛凝胶贴膏外敷,给药4周后,用Aloka Latheta LCT-200观察兔膝骨关节影像学改变、用ELISA法检测血清IL-1、TNF-α、MMP-13、TIMP-1含量。结果 C组兔膝关节病变程度较B组轻,C组兔血清中IL-1、TNF-α、MMP-13含量较B组降低(P<0. 05),TIMP-1含量较B组升高(P <0. 05)。结论南星骨痛凝胶贴膏能减轻兔膝关节骨关节炎病变程度,对关节软骨有保护作用,其作用机制可能与抑制炎症因子IL-1、TNF-α及MMP-13表达,增加TIMP-1表达有关。

Effect of Nanxing Gutong Ointment on Joint Morphology and Serum Levels of IL-1,TNF-α,MMP-13 and TIMP-1 in Rabbit Knee Osteoarthritis

Objective To examine the anti-inflammatory mechanism of Nanxing Gutong Ointment on treating knee osteoarthritis by evaluating its effects on levels of IL-1,TNF-α,MMP-13 and TIMP-1in peripheral blood and joints imaging changesin papain-inducedknee osteoarthritis rabbits.Methods Experimental study:18 New Zealand rabbits were randomly divided into 3 groups:blank group(A),model group(B),Nanxing Gutong Ointment external application treatment group(C),with 6 rabbits in each group.The KOA rabbit model was created(except the blank group)through the knee joint intracavitary injection of papain.Then the rabbits were treated for 4 weeks(28 days).After the treatment period,the blood was obtained fromear vein to examine the levels of IL-1,TNF-α,MMP-13 and TIMP-1.Then the experimental rabbits were sacrificed by air embolization.One rabbit was randomly selected from each group and the knee joint was taken to observe the joints imaging changes.Results 1.The results of joints imaging showed that the degree of joint disease in the treatment group was lighter than that in the model group.2.Comparison between the model group and the blank group:The statistically significant difference(P<0.05)was observed for levels of IL-1,TNF-α,MMP-13 and TIMP-1 in the rabbit peripheral blood.After the treatment period of 28 days,the statistically significant difference(P<0.05)was also observed between the treatment groupand the model group.Conclusion External application of Nanxing Gutong Ointment can inhibit the expressions of inflammatory cytokines IL-1 and TNF-αin the rabbit peripheral blood,inhibit the expression of MMP-13,increase the expression of TIMP-1,protect the articular cartilage,improve symptoms related to knee osteoarthritis and delay the course of the disease.
