银杏内酯B对胆固醇和载脂蛋白E4损伤海马神经元凋亡的保护作用

摘    要:目的:探讨银杏内酯B对高胆固醇和apoE4致海马神经元凋亡的保护作用。方法:采用无血清培养基体外培养新生大鼠海马神经元,采用神经元特异性烯醇化酶免疫萤光进行鉴定。用160μg/mL银杏内酯B处理海马神经元16h,40μg/mL 25-OH-胆固醇和30μg/mL载脂蛋白E4继续处理24h。通过Hoechst试剂盒测定神经元的凋亡,实时定量RT-PCR测定bax和Bcl-2 mRNA的表达。结果:银杏内酯B抑制神经元凋亡,上调bcl-2 mRNA的表达,下调bax mRNA的表达。结论:银杏内酯B对高胆固醇和apoE4损伤海马神经元凋亡有保护作用,其机理可能与调节两种凋亡相关基因有关。

Abstract:Objective:To observe the effect of Ginkgolide B on the apotosis of cultured hippocampi neurons exposed to cholesterol and apoliptein E4.Methods:Newborned rat hippocampi nrurons were cultured with serum-free medium and identified with enolase by immunofluorescence.Hippocampi nrurons were incubated with 160μg/mL Ginkgolide B for 16h and then 40μg/mL 25-OH-cholesterol and 30 μg/mL apoE4 for 24h.The apotosis of hippocampi nrurons was detected by Hoechst 33258 kit,the mRNA expression of Bax and Bcl-2 was detected by real time quantitative RT-PCR.Results:Ginkgolide B can markedly inhibite apotosis of neurons.The data also showed that bcl-2 mRNA expression was upregulated as well as bax mRNA expression was downregulated in Ginkgolide B group.Conclusions:Ginkgolide B had effect on hippocampi neurons apotosis induced by cholesterol and apoE4,the effect may be related to regulate the apotosis relative gene bcl-2 and bax mRNA expression.
