摘    要:目的:观察罗勒多糖对人卵巢癌细胞SKOV3细胞骨架的影响,探讨罗勒多糖抗卵巢癌细胞侵袭转移的可能机制。方法:将卵巢癌SKOV3细胞分为4组,A组为不加罗勒多糖处理的空白对照组,B、C、D组分别用不同浓度罗勒多糖处理,以罗丹明-鬼笔环肽标记微丝,免疫荧光法标记微管,用荧光显微镜观察。结果:罗勒多糖作用的SKOV3细胞微丝重排、伪足减少、微管蛋白弥散分布、细胞骨架的网络结构发生改变;这一作用与剂量呈正相关。结论:罗勒多糖可能通过影响人卵巢癌SKOV3细胞微丝、微管来调节其运动性和侵袭力。

Abstract:Objective:To observe the effect of the basil polysaccharides on ovarian cancer SKOV3 cytoskele- ton,and reveal the mechanism of basil polysaccharide to inhibit the metastasis of ovarian cancer. Methods: The SKOV3 cells were divided into 4 groups. Group A was the blank control group without basil polysac- charides;different concentrations of basil polysaccharide were used to treat the cells of B,C,D groups;group E was positive control group with taxol;Rhodamine-phaltoidin was used to mark the microfilaments,the mi- crotubules was labeled by immunofluorescence and then observed by fluorescence microscopy. Results:The microfilaments of the cells which treated by basil polysaccharides were rearranged and the pseudopodia was reduced. The microtubules were more dispersive and the network structure of the eytoskeleton changed at the same time. Also there is a positive correlation between effect and dosage. Conclusion:Basil polysaccharide can regulate the motility and invasiveness of the SKOV3 cells by influencing their microfilaments and micro- tubules.
