基于网络药理学方法赤芍-牡丹皮药对治疗中风作用机制探讨

摘    要:目的:采用网络药理学的分析方法研究赤芍-牡丹皮经典药对治疗中风的分子作用机制。方法:基于中药系统药理数据库和分析平台(TCMSP)、药物与疾病数据库(Drug Bank)、赤芍-牡丹皮的疾病与化学成分靶点网络则分别由治疗靶标数据库(TTD)建立,而它的药效成分与靶点网络主要运用Cytoscape 3.7.0软件构建而成,并同时进行了网络拓补分析; 至于核心靶点对基因本体(GO)的功能富集则主要运用DAVID数据库完成,同时也对百科全书KEGG(基因组)通路的功能富集进行了研究。结果:网络拓补分析结果表明,最核心的药效成分可能为槲皮素、山奈酚、鞣花酸、黄芩苷,最核心的GO功能有活性氧代谢过程、脂多糖、核受体活性、细胞因子受体结合、细胞膜微区、囊腔作用等; 最核心的作用通路有VERF通路、Toll样受体通路、肿瘤坏死因子(TNF)通路、白细胞介素-17(IL-17)信号通路、磷脂酰肌醇3激酶-蛋白激酶b(P13K-Akt)通路等。结论:本文基于网络药理学方法对赤芍-牡丹皮药对干预中风的作用机制进行了分析发现,赤芍-牡丹皮药对可能是通过VERF通路、Toll样受体通路、TNF通路、IL-17信号通路、P13K-Akt等通路协同作用发挥治疗缓解中风功效,上述预测结果可为赤芍-牡丹皮药对在中风治疗的药理作用研究及临床应用提供一定的理论依据。

Abstract:Objective:The molecular mechanism of “Radix paeoniae rubra-Cortex moutan pair” in the treatment of stroke was studied by network pharmacology.Methods:The target networks of diseases and chemical components based on TCMSP,Drug Bank and “Radix paeoniae rubra-Cortex moutan pair” were established by TTD respectively,and its pharmacodynamic components and target networks were mainly constructed by Cytoscape 3.7.0 software.As for the functional enrichment of Gene Ontology(GO)by core targets,david database is mainly used.At the same time,the functional enrichment of KEGG(genome)pathway in encyclopedia is also studied.Results:The results of network complementation analysis showed that the core pharmacodynamic components may be quercetin,kaempferol,ellagic acid and baicalin.The core GO functions include reactive oxygen species metabolism,lipopolysaccharide,nuclear receptor activity,cytokine receptor binding,cell membrane microdomain,vesicle effect and so on; IL-17 kinase is the most important signaling pathways of tumor cells.Conclusion:Based on the network pharmacology method,this paper analyzes the mechanism of “Radix paeoniae rubra-Cortex moutan pair” on the intervention of stroke.It is found that radix paeoniae rubra cortex moutan pair may play the role of treating and alleviating stroke through the synergistic effect of verf pathway,toll like receptor pathway,TNF pathway,IL-17 signaling pathway and P13K-Akt pathway.The above prediction results can provide a theoretical basis for the pharmacological study and clinical application of “Radix paeoniae rubra-Cortex moutan pair” in the treatment of stroke.
