加味大黄?虫颗粒对精索静脉曲张模型大鼠精子线粒体结构的影响

摘    要:目的:探索加味大黄?虫颗粒对精索静脉曲张模型大鼠精子线粒体结构的影响。方法:40只雄性大鼠随机选出10只作为假手术组,剩余30按照Turner方法制造左侧精索静脉曲张模型大鼠,造模完成后,按照随机数字表法分为加味大黄?虫颗粒组、模型组、迈之灵组,每组各10只。加味大黄?虫颗粒组给予加味大黄?虫颗粒灌胃; 迈之灵组予迈之灵片,假手术组和模型组分别予等剂量0.9%生理盐水灌胃。8周后处死所有大鼠,取出左侧睾丸组织,完成精液质量分析、睾丸组织病理检测。结果:灌胃8周后,加味大黄?虫颗粒组精子浓度、总活率、前向运动精子、精子直线速度(VSL)、精子曲线速度(VCL)、平均路径速度(VAP)、精子头侧摆幅度(ALH),明显高于模型组(P<0.05)。模型组睾丸组织病理报告生精小管管腔空化,各级生精细胞破坏严重,精子膜结构破坏,精子线粒体肿胀、溶解。而加味大黄?虫颗粒组中睾丸组织的病理损伤得到不同程度的改善,精子线粒体结构完整,形态大小规则。结论:加味大黄?虫颗粒在一定程度上修复精索静脉曲张大鼠睾丸组织的病理损伤,进而起到保护精子线粒体结构与功能的作用,为精子的发育、活动提供稳定的能量代谢环境,有助提高精子活力及增加受精能力。

Abstract:Objective:To explore the effect of modified Dahuang Zhechong granule on sperm mitochondria structure in varicocele model rats.Methods:10 of 40 male rats were randomly selected as sham operation group,and the remaining 30 left varicocele model rats were made according to Turner method.After the model was completed,they were divided into modified Dahuang Zhechong granule group,model group and Maizhiling group according to random number table method,with 10 rats in each group.In the modified Dahuang Zhechong granule group was made into granules with a concentration.Maizhiling group was given Maizhiling tablets,and sham operation group and model group were given equal amount of 0.9 saline by gavage.8 weeks later,kill all the rats,taken out the left testis tissue,complete semen quality analysis and testicular histopathological examination.Results:After 8 weeks of intragastric administration,sperm concentration,total viability,forward moving sperm,sperm linear velocity(VSL),sperm curve velocity(VCL),average path velocity(VAP),sperm lateral swing amplitude(ALH)in the modified Dahuang Zhechong granule group were significantly higher than those in the model group(P<0.05).Testicular histopathology in the model group reported that the seminiferous tubules were cavitated,the spermatogenic cells at all levels were seriously damaged,the sperm membrane structure was damaged,and the sperm mitochondria were swollen and dissolved.However,the pathological damage of testis tissue in the modified rhubarb vermilion granule group was improved to varying degrees,and the structure of sperm mitochondria was complete and the shape and size were regular.Conclusion:Modified Dahuang Zhechong granules can repair the pathological damage of testicular tissue in varicocele rats to a certain extent,provide a stable energy metabolism environment for sperm development and activity,and help improve sperm motility and fertilization ability.
