六味补气方对COPD模型大鼠Th1/Th2类细胞平衡调节作用的实验研究

摘    要:目的:研究六味补气方对慢性阻塞性肺疾病(COPD)模型大鼠气道炎症及免疫功能的影响及其作用机制。方法:将SD大鼠按随机数字表法分为正常对照组、模型组、桂龙咳喘宁组和六味补气方组,每组10只。除正常对照组外,其余各组采用烟熏加脂多糖(LPS)气管滴入方法建立COPD实验动物模型,造模成功后,分别给予0.9%氯化钠溶液及相应药物灌胃,连续干预14 d。于造模第4周和药物干预结束后检测肺功能; 苏木精-伊红(HE)染色法光镜观察肺组织的病理学改变; 酶联免疫吸附法(ELISA)检测血清及肺泡灌洗液(BALF)中白细胞介素-4(IL-4)、干扰素-γ(IFN-γ)水平,并分析辅助性T细胞Th1/Th2平衡状态。结果:与正常对照组比较,模型组大鼠出现COPD症状和体征,肺功能降低,血清及BALF中IFN-γ水平明显下降,IL-4水平明显增加(均P<0.05),Th1/Th2平衡向Th2方向移动; 与模型组相比,六味补气方组和桂龙咳喘宁组大鼠一般情况及肺功能均有不同程度改善,血清及BALF中IFN-γ水平明显增加,IL-4水平下降(均P<0.05); 与桂龙咳喘宁组相比,六味补气方组大鼠COPD症状体征减轻,血清及BALF中IFN-γ水平明显增加,IL-4水平下降(均P<0.05),Th1/Th2平衡向Th1方向移动。结论:六味补气方可改善COPD模型大鼠肺功能,减轻气道炎症,其机制可能是通过调节Th1/Th2类细胞平衡,增加免疫功能实现的。

Abstract:Objective:To study the effects of Liuwei Buqi recipe on airway inflammation and immune function in rats with chronic obstructive pulmonary disease(COPD)model and its mechanism of action.Methods:SD rats were divided into normal control group,model group,Guilong Kechuanning group and Liuwei Buqi recipe group according to the random number table method,with 10 rats in each group.Except for the normal control group,the experimental animal models of COPD were established by smoked plus lipopolysaccharide(LPS)tracheal drip method,and after successful modeling,saline and corresponding drugs were given by gavage for 14 d.Lung function was measured at the fourth week of modeling and at the end of drug intervention;pathological changes in lung tissue were observed by light microscopy using hematoxylin-eosin(HE)staining;IL-4 and IFN-γlevels in serum and alveolar lavage fluid(BALF)were measured by enzyme-linked immunosorbent assay(ELISA),and the Th1/Th2 was analyzed.Results:Compared with the normal control group,rats in the model group showed signs and symptoms of COPD,reduced lung function,significantly decreased IFN-γlevels in serum and BALF,significantly increased IL-4 levels(P<0.05),and shifted Th1/Th2 balance toward Th2;compared with the model group,rats in the Liuwei Buqi recipe group and Guilong Kechuanning suspension group showed different degrees of improvement in general condition and lung function,and significantly increased IFN-γlevels in serum and BALF,and decreased IL-4 levels(all P<0.05).Compared with the model group,the general condition and lung function of the rats in the Guilong Kechuanning suspension group and Guilong Kechuanning group showed different degrees of improvement,and the levels of IFN-γin serum and BALF increased significantly,while the levels of IL-4 decreased significantly(all P<0.05);compared with the Guilong Kechuanning group,the symptoms and signs of COPD in the rats in the Guilong Kechuanning group decreased,and the levels of IFN-γin serum and BALF increased significantly,while the levels of IL-4 decreased significantly(all P<0.05).Conclusion:Liuwei Buqi recipe can improve lung function and reduce airway inflammation in rats with COPD model,and the mechanism may be achieved by regulating the balance of Th1/Th2-like cells and increasing immune function.
