醋柳黄酮单体对高密度脂蛋白氧化修饰的影响

摘    要:目的 观察醋柳黄酮主要单体槲皮素 (Que)和异鼠李素 (Iso)在体外对Fe2 + 介导的人血浆高密度脂蛋白 (HDL)氧化修饰的影响。方法 采用一次性密度梯度法分离正常人血浆HDL ,用Fe2 + 进行体外氧化修饰。抗氧化组在温育前加入不同浓度Que或Iso(5 0~ 2 0 0μmol/L) ,分别检测脂蛋白中丙二醛 (MDA)、维生素E含量及超氧化物歧化酶 (SOD)活性。结果Que和Iso可明显减少氧化型HDL(OX -HDL)中MDA生成 (P <0 .0 0 1) ,并呈现一定的浓度效应 ;同时可明显延缓维生素E含量降低周期 ,显著提高SOD活性 (P <0 .0 0 1)。结论 醋柳黄酮主要单体Que和Iso均对Fe2 + 介导的HDL氧化修饰具有明显的抑制作用 ,其作用途经可能与抗氧化自由基有关 ,该类物质可能是一种良好的抗氧化剂。

Abstract:Objective It is to observe the effects of quercetin ( Que) and isorhamne tin (Iso) which are the main hippophae rhamnoides flavonoid monomers on the oxid ative modification of human plasma high density lipoprotein (HDL) mediated by Fe 2+ in vitro.Methods Human plasma HDL was segregated by one-step density gradi ent separation and oxidized modification with Fe 2+ in vitro. Que and Iso in different con centra tion (50~200 μmol/L) were added in anti-oxidative group before incubation. Th e contents of malondialdehyde (MDA) and vitamin E in lipoprotein and the activity of superoxide dismutase (SOD) were detected respectively.Results Que and Iso could significantly reduce MDA producti on in oxidized HDL (OX-HDL) (P<0.001), and it showed definite concentr ation effect. Que and Iso c ould evidently postpone vitamin E content depressing period and markedly elevate SOD activity (P<0.001).Conclusion Que and Iso which are the main hippophae rhamnoides fla vonoid monomer s have obvious inhibit effect on the oxidative modification of HDL mediated by F e 2+. The action pass may be correlated with anti-oxidative free radical. T he kind of material may be a satisfactory antioxidant.
