类风湿关节炎患者血清sFas/sFasL及TNFα的变化及相关性研究

摘    要:目的 :探讨 s F as与 s F as L和 TNFα在类风湿关节炎 ( RA )发病中的意义。方法 :采用固相酶联免疫分析( EL ISA)法检测 38例 RA患者血清 s Fas,s F as L含量及 TNFα水平。结果 :RA患者血清 s F as,s Fas L ,TNFα水平均高于正常对照组 ,RA患者活动期 s F as L,TNFα显著高于非活动期 ( P<0 .0 1) ,而 s Fas水平未见显著差异。在 38例 RA患者中 ,有 13例 s F as和 s F as L 的水平同时升高 ,RA活动期患者血清 s Fas L 的增高与 TNFα增高呈正相关 ( r=0 .46 9P<0 .0 1) ,而其它组间无相关性 ( P>0 .0 5 )。结论 :s Fas L水平与 RA的炎症程度有关 ,高浓度 s Fas和 s F as L可抑制 Ts细胞对 Th的负向调节。TNFα与 s Fas L具有关联性。血清 s Fas,s Fas L ,TNFα检测有助于对 RA病情进展作出判断 ,并为 RA的免疫生物治疗提供新方法。

Abstract:Objective:It is to investigate the role of soluble Fas(sFas), soluble Fas ligand (sFasL), TNFα in pathogeny of RA. Methods: The levels of sFas, sFasL, TNFα in serum of thirty-eight patients with RA were detected by enzyme-linked immunosorbent assay(ELISA). Results: The levels of sFas, sFasL, TNFα in serum of thirty-eight patients with RA were higher than those of healthy individuals. The serum levels of sFasL, TNFα in RA active period were significantly higher than those in inactive period(P<0.01). The levels of sFas, sFasL were both increased in thirteen RA patients. The serum levels of sFasL in patients with RA in active period were correlated with TNFα (r=0.469, P<0.01). Conclusion:The serum levels of aFasL were positively related to inflammatory degree of RA patients. High serum levels of sFas and sFasL could suppress downregulation of Ts to Th lymphocytes. TNFα was related to sFasL. The measurement of sFas, sFasL, TNFα is helpful to know the RA patient's condition and supply a bio-immunological treatment for the RA patient. ;
