卡维地洛对血管紧张素Ⅱ介导的心脏成纤维细胞Smad蛋白表达的影响

目的应用血管紧张素Ⅱ(AngⅡ)刺激心脏成纤维细胞,观察卡维地洛(carvedilol)对Smad蛋白表达的影响。方法carvedilol干预体外培养的大鼠心脏成纤维细胞,用替米沙坦(telmisartan)和美托洛尔(metoprolol)作对比,应用Western blot方法检测AngⅡ诱导的心脏成纤维细胞Smad2、磷酸化Smad2(p-Smad2)和Smad4的表达情况。结果与空白对照组相比较,AngⅡ刺激组Smad2、p-Smad2表达显著增高(P均<0.01),Smad4的表达也明显增加(P均<0.05);carvedilol和telmisartan干预后可显著减低Smad2、p-Smad2的表达(P<0.01),telmisartan还能抑制Smad4的表达(P<0.01);而metoprolol对Smad的表达没有明显影响。结论carvedilol能抑制AngⅡ介导的Smad蛋白在心脏成纤维细胞中的表达。

Effect of carvedilol on expression of Smad proteins mediated by AngⅡin cardiac fibroblasts

Objective It is to investigate the effect of carvedilol on the expression of Smad proteins mediated by Ang Ⅱ in cardiac fibroblasts.Methods Primary cardiac fibroblasts were dissociated from ventricles of Sprague-Dawley(SD) rats after stimulated with AngⅡ,telmisartan and metoprolol were used as control.The expression of Smad2,p-Smad2 and Smad4 were detected with Western blot.Results Compared with control group,the expression of Smad2,p-Smad2 and Smad4 were obviously raise mediated by AngⅡ(P<0.01 or P<0.05).Both carvedilol and telmisartan could significantly reduced the expression of Smad2 and p-Smad2(P<0.01),but telmisartan could reduced Smad4.Meanwhile,metoprolol did not show any effect on the synthesis of all the Samds after AngⅡ stimulation.Conclusion Carvedilol can reduce the expression of Smad proteins mediated by AngⅡ in cardiac fibroblasts.
