金钗石斛提取物抗白内障的体外实验研究

摘    要:目的研究金钗石斛总生物碱和粗多糖对白内障的抑制作用。方法采用溶剂提取法分离提取金钗石斛总生物碱和粗多糖;DMEM低糖培养基体外培养Wistar大鼠晶状体,将晶状体分为7个组,分别在培养后6、12、18、24 h四个时段观察并记录晶状体混浊度的改变;最后,检测各组晶状体水溶性蛋白、GSH的含量、及T-SOD和MDA的活性。结果在培养的各个时段,正常对照组晶状体始终保持透明,各药物组晶状体混浊度与模型组相比明显较轻(P〈0.05),而石斛总生物碱高剂量组和阳性对照组混浊程度最轻;金钗石斛总生物碱组、粗多糖组晶状体水溶性蛋白、GSH含量及T-SOD活性均升高,同时MDA的活性降低;其中总生物碱高剂量组晶状体水溶性蛋白含量显著高于阳性对照组水平(P〈0.05),其MDA活性则显著低于阳性对照组(P〈0.05)。结论金钗石斛总生物碱和粗多糖在体外均有一定的抗白内障之效,而总生物碱的效果优于粗多糖。

Abstract:Objective To probe into the inhibitory effects of the total alkaloids and polysaccharides on cataract lens in vitro from extracted of Dendrobium nobile.Methods The total alkaloids and polysaccharides were extracted from Dendrobium nobile;Wistar rats' crystalline lenses were cultured in DMEM medium and divided into 7 groups. The opacity of each crystalline lens was observed and recorded respectively with different time.Finally,MDA of each lens was measured with the kits respectively.Results The content of water-solubility protein and GSH and the activity of T-SOD were increased in the groups of total alkaloids and polysaccharides,and the activity of MDA was decreased as well.Conclusion The total alkaloids and polysaccharides of Dendrobium nobile had both inhibited cataract lens which caused by oxidative damage in vitro, and the effect of total alkaloids was better than that of polysaccharide.
