芪蛭益肺颗粒对COPD模型大鼠肺组织中气道重塑相关蛋白的影响

摘    要:目的探究芪蛭益肺颗粒对COPD模型大鼠气道重塑抑制作用的作用机制。方法使用自制烟熏箱烟熏大鼠,持续烟熏28 d,于第1、11、21天气道内滴注LPS,滴药日不熏烟。实验第21天起,各组大鼠每日灌胃给与相应药物一次,连续给药40 d。免疫组化法测定肺组织中MMP-9、MMP-12及TIMP-1的蛋白表达水平。结果芪蛭益肺颗粒中、低剂量组MMP-9的表达显著下降(P <0.05);芪蛭益肺颗粒高、低剂量组MMP-12表达显著下降(P <0.05);罗红霉素组、芪蛭益肺颗粒高、低剂量组TIPM-1的表达显著下降(P <0.05)。结论芪蛭益肺颗粒可通过调节金属蛋白酶-抗金属蛋白酶体系的动态平衡,产生抑制COPD模型大鼠气道重塑的作用。

Effects of Qizhi Yifei Granules on Expression Level of Airway Remodeling Associated Proteins in Lung Tissue of COPD Rats

Abstract:Objective To investigate the mechanism of Qizhi Yifei Granules on airway remodeling in COPD rats. Method Wistar rats are randomly divided into 6 groups: vehicle control group, model control group, roxithromycin group, and three dosage groups of Qizhi Yifei Granules. A rat model of COPD is established by infusion LPS with cigarettes smoke exposure. Rats exposed to smoke once a day for 28 days.LPS is injected into the airways on the 1 st, 11 th and 21 st day. From the 21 st day of the experiment, each group rats are administered the specific drugs once a day for 40 days. On the 21 st day, the expression levels of MMP-9, MMP-12 and TIMP-1 in lung tissue are measured with immunohistochemistry. Results Compared with the model group, the expression levels of MMP-9 are significantly decreasedin the groups of Qizhi Yifei granules at 9.10 and 4.55 g/kg(P < 0.05). The expression levels of MMP-12 are significantly decreased in the groups of Qizhi Yifei granules at 18.20 and 4.55 g/kg(P < 0.05). The expression level of TIMP-1 is significantly decreased in the roxithromycin group and groups of Qizhi Yifei granules at 18.20 and 4.55 g/kg(P < 0.05). Conclusion Qizhi Yifei Granules could inhibit airway remodeling in COPD rats by regulating the homeostasis of the metalloproteinase-antimetalloproteinase system.
