增液汤对干燥综合征模型鼠血清TNF-α和IL-1β影响的研究

摘    要:目的:观察增液汤对干燥综合征模型鼠血清中TNF-α和IL-1β的影响.方法:54只ICR小鼠随机分为6组,即空白组、模型组、来氟米特组和增液汤高、中、低剂量组.采用弗氏完全佐剂及ICR小鼠颌下腺蛋白抗原联合诱导SS动物模型,60天后眼球取血,采用ELISA法测定各组小鼠血清中TNF-α和IL-1 β的含量.结果:模型组小鼠血清中TNF-α和IL-1β的含量均高于空白组,有显著性差异(P<0.01);来氟米特组和增液汤高、中、低剂量组小鼠血清中TNF-α和IL-1β的含量均低于模型组,差异显著(P<0.01),增液汤高剂量组小鼠血清中TNF-α和IL-1β的含量明显低于来氟米特组(P<0.01),中剂量组小鼠血清中TNF-α和IL-1β的含量介于高、低剂量组之间.结论:增液汤治疗SS模型小鼠的机制之一可能是降低了血清中TNF-α和IL-1β的表达水平,且增液汤剂量越大效果越明显.

Effect of Zengye Decoction on Serum TNF-α and IL-1β of Model Rats with Sjogren Syndrome

Abstract:Objective : To observe the mechanism of decoction for serum TNF - alpha and IL - 1 beta of model rats with sjogren syndrome. Methods :54 ICR mice were divided into 6 groups randomly: normal control group, model group, Leflunomide group, Zengye decoction high dose, middle dose and low dose groups. Develop the mice models with sjogren syndrome by injecting FCA combined with protein antigen of the submandibular gland of ICR mice. 60 days later by the eyeball blood, determine serum levels of TNF - alpha and IL - 1 beta by ELISA method. Results: The contents of serum TNF - alpha and IL - 1 beta in model group were higher than those in the blank group,and there was a significant difference (P 〈0.01 ) ;as to the Leflunomide group and Zengye decoction high, low dose groups, the contents of serum TNF - α and IL - 1 In were lower than those in the model group, and the difference was significant (P 〈 0.01 ). In decoction high dose group the serum levels of TNF - alpha and IL - 1 beta were significantly lower than those in the leflunomide group ( P 〈 O. O1 ). In middle dose group TNF - α and IL - 1 In were significantly between high and low dose groups. Conclusion: One of the mechanisms of Zengye decoction in the treatment of SS mice may be that it decreases the expression of serum levels of TNF - alpha and IL - 1 beta, and the more the Zengye decoction is, the more obvious its effect is.
