黄芪含药血清对H2O2诱导人脐静脉内皮细胞氧化损伤模型细胞活性的影响

摘    要:目的:观察养心汤方中单味中药黄芪不同浓度含药血清对H:O:诱导人脐静脉内皮细胞(HU—VEC)氧化损伤模型细胞增殖的影响及对损伤HUVEC的保护作用,以进一步研究养心汤对冠心痛不稳定型心绞痛的确切作用机制。方法:以浓度为1.03g·mL^-1的黄芪药液灌胃大鼠1周,于末次给药后取血离心制备含药血清。将HUVEC与不同剂量浓度(2%、4%、6%)单味黄芪含药血清、基础培养液、空白血清培养液一起培养,共分5组,培养24h后加入H2O2作用2h,通过MTT法及形态学方法观察不同浓度黄芪含药血清对H2O2损伤HUVEC生长活性变化。结果:黄芪含药血清2%、4%、6%各组细胞OD值均较H2O2模型对照组明显升高,与H2O2模型组相比较均有统计学意义(P〈0.01),且2%、4%、6%组间比差异有统计学意义(P〈0.05)。形态学方面,黄芪含药血清可使细胞体积变小,间隙变大,但彼此之间存在联系,脱落细胞较少;皱缩的程度明显改善。结论:黄芪含药血清可以促进H2O2损伤人脐静脉内皮细胞的增殖,增强HUVEC抗氧化损伤能力,逆转和改善内皮功能障碍,具有一定剂量依赖关系。

Abstract:Objective :Yang Xin Tang Fong single herb Astragalus serum contains different concentrations of H2O2 induced human umbilical vein endothelial cells (HUVEC) oxidative damage model of cell proliferation and the protective effect of HUVEC on the injury to further research to support Coronary heart disease with un stable angina exact mechanism of action. Methods: week after the last administration blood centrifugation Astragalus liquid concentration 1.03g·mL^-1 fed rats 1 preparation containing serum. The concentration of HUVEC with different doses (2% ,4% ,6% ) single Chinese Astragalus containing serum, basal medium, control serum with culture medium, divided into 5 groups, the role of culture 24h after adding H2O2 2h, by MTF method and morphological effects of different concentrations of Astragalus containing serum on the activity of H2O2 injury in HUVEC growth. Results:Astragalus containing serum 2% ,4% ,6% OD values in each group which was compared with model control group was significantly higher H2O2, and H2O2 model group was statistically significant compared ( P 〈 0. 01 ), and 2%, 4%, 6% group was significantly higher than ( P 〈 0. 05 ). Morphological, Astragalus containing serum induced an smaller, space becomes larger, but there is a link between each other, less exfoliated cells ; significant improvement in the degree of shrinkage. Conclusion : Astragalus containing serum can promote H2O2 injury in enhance the ability of HUVEC against oxidative damage human umbilical vein endothelial cell proliferation, reversal, improve endothelial dysfunction, and has a dose - dependent.
