肾虚血瘀-子宫内膜容受性不良模型大鼠的建立

摘    要:目的:建立肾虚血瘀一子宫内膜容受性不良大鼠模型,为研究补肾活血法改善子宫内膜容受性的作用机理提供病证结合实验动物模型。方法:将48只SD大鼠随机分为模型组、阳性对照组及空白组,每组各16只。模型组予羟基脲450mg·(kg·d)-1,灌胃10d,第4天开始加用肾上腺素0.3mg·(kg·d)-1皮下注射7d,阳性对照组(胚胎着床障碍动物模型)、空白纽予等量生理盐水灌胃10d,第4天开始加用等量生理盐水皮下注射7d,建立大鼠模型。结果:模型组高、中、低切变率下全血黏度、聚集指数与空白组、阳性对照组比较,均明显升高(P〈0.05)。与空白组比较,模型组及阳性对照组大鼠透射电镜下子宫内膜上皮细胞核结构不规则,核内异染色质增多,线粒体数量减少,空泡化,内质网水肿扩张明显,大小不均;扫描电镜下胞饮突数量明显减少,发育差,分布不均;而模型组与阳性对照组均无明显差异。妊娠第8天,模型组、阳性对照组的胚泡着床率和平均着床点数均比空白组大鼠明显减少(P〈0.05)。而模型组与阳性对照组无明显差异。结论:羟基脲灌胃450mg·(kg·d)-1,造模10d,第4天加用肾上腺素0.3mg·(kg·d)-1皮下注射7d,可成功建立肾虚血瘀一子宫内膜容受性不良大鼠模型。

Abstract:Objective:To establish the model of rats with poor endometrial receptivity with blond stasis due to kidney deficiency, and provide the experimental animal model of integration of disease and syndrome for the research into the fhnetional mechanism of improving the endometrial reeeptivity by using nourishing- kidney -activating- blood method. Methods:48 SD rats were ran- domly divided into the model group,the positive control group(rats were injected with mifepristone),and the blank group( the nor- real control group) with 16 in each group. The rats' stomach in the model group were td with Hydroxyurea 450 mg ( kg d ) - for ten days continuously;at the same time,from the 4th day,they were added ad,'enaline 0.3 mg (kg d) l in the form of subcu- taneous injection for 7 days. The rats' stomach in the positive control group( embryo implantation barrier animal model) and in the blank were fed with the same amount of normal saline for 10 days,and at the same time from the 4th day,they were added the e- qual amount normal saline in the form of subcutaneous injection for 7 days. In this way,the required rat model was estalished. Results:Compared with the blank group and the positive contrnl group,in the model group,the whole blood viscosity and aggrega- tion all obviously rose in spite of high,middle or low shear rate(P 〈 0.05 ). Compared with the blank group, in the mode group and in the positive control groui'),observed through transmission electron microscope,the epithelial nuclei stncture of rats' emto- metrium was irregular; The amount of heterochrumatin inside the nuclei Jose ; The amount of tnilochoudria decreased, and there was cell vacuolation ; And endoplasmic reticulum swelling was obviously expanded with uneven size ;Scanning electron mi'mseope, the amount of pinopodes with poor development and uneven distribution obviously decreased, but there was no clear dift-'rence be- tween the model grnup and the positive contrx)l group. 8 days later" alter pregnancy, in the model group and the positive control group,both the blastocyst implantation rate and the average bedding points obviously decreased c,ompared with the rats in the blank group( P 〈0.05). Conclusion :10 days later after rat's stomach was fed with hydroxyurea 450 mg (kg d) -, and at the same time, from the 4'h day, rats were added adrenaline 0.3 mg ( kg d) -t in the form of subeutmleus injection tot 7 days,the model of rats' poor endometrial reeeptivity with blood stasis due to kidney deficiency can be successfidly established.
