川芎嗪预处理对大鼠心肌缺血再灌注损伤作用及其机制研究

摘    要:目的:探讨川芎嗪预处理对大鼠心肌缺血再灌注损伤的保护作用。方法:将30只Wistar大鼠随机分为3组:假手术组、缺血再灌注损伤组、川芎嗪预处理组。建立大鼠心肌缺血再灌注损伤模型,测定SOD、GSH-Px活性,应用原位杂交技术和免疫组化S-P法检测HSP70蛋白的表达。结果:与缺血再灌注损伤组比较,川芎嗪预处理组SOD(P<0.01)、GSH-Px(P<0.05)活力显著升高,HSP70mRNA(P<0.01)及其蛋白(P<0.05)的表达显著增加。结论:SOD、GSH-Px和HSP70是心肌中重要的内源性保护物质,川芎嗪可增加SOD、GSH-Px活性,并诱导HSP70mRNA及其蛋白的表达而发挥心肌保护作用。

Abstract:Objective:To probe into the protective effect of chuanxiongzine pretreatment on rats' myocardial ischemia reperfusion injury.Methods:30 Wistar rats were randomly divided into 3 groups:sham operation group,the group of ischemia reperfusion injury and the group of chuanxiongzine pretreatment.The model rats with myocardial ischemia reperfusion injury was built and SOD and GSH-Px activity were detected.In situ hybridization and Streptavidin-peroxidase(S-P) immunohistochemical method were used to detect the expression of heat shock protein 70.Results:Compared with the group of ischemia reperfusion injury,the group of chuanxiongzine pretreatment had an evident increase of vatality:SOD(P<0.01) and GSH-Px(P<0.05),and a remarkably increased experssion of HSP70 mRNA(P<0.01) and protine(P<0.05).Conclusion:SOD,GSH-Px and HSP70 were the important endogenous protective substances in the myocardium.Chuanxiongzine can enhance the activity of SOD and GSH-Px and induce the HSP70mRNA and the expression of its protein,which was the protective effect of myocardium.
