白芍总苷对急性肝损伤小鼠肝脏保护作用的研究

摘    要:目的:探讨白芍总苷(total glucosides of paeony,TGP)对四氯化碳(CCl4)致急性肝损伤小鼠肝脏的保护作用。方法:90只小鼠按体质量随机分为正常对照组、模型对照组、TGP高剂量组、TGP中剂量组、TGP低剂量组和双环醇片组,每组各15只。TGP各剂量组自造模前1周起,以15μL·g-1为给药剂量灌胃,正常对照组和模型对照组给予等容量生理盐水,双环醇片组给予110 mg·kg-1双环醇片溶液灌胃,每天1次,连续7 d。建立CCl4诱导小鼠化学性肝损伤模型,造模24 h后自小鼠后眼眶静脉丛采血0.4 mL,使用试剂盒测定法检测中血清丙氨酸氨基转移酶(alanine transaminase,ALT)、天门冬氨酸氨基转移酶(aspartate aminotransferase,AST)活性;采用Lowry法检测肝匀浆中丙二醛(malondialdehyde,MDA)、超氧化物歧化酶(superoxide dismutase,SOD)、谷胱甘肽过氧化物酶(glutathione peroxidase,GPX)的组织蛋白含量;HE染色小鼠肝组织作病理检查并进行分析。结果:模型对照组小鼠血清ALT、AST水平高于正常对照组,TGP各剂量组及双环醇片组血清ALT、AST水平低于模型对照组,差异均有统计学意义(P<0.05)。模型对照组小鼠肝匀浆MDA水平高于正常对照组,SOD、GPX含量低于正常对照组,差异具有统计学意义(P<0.05)。TGP各剂量组MDA水平低于模型对照组,SOD、GPX含量高于模型对照组,差异具有统计学意义(P<0.05);双环醇片组MDA水平低于模型对照组,差异具有统计学意义(P<0.05)。结论:白芍总苷对CCl4致小鼠急性肝损伤有确切的保护作用,其作用机制与调节免疫、抗氧化密切相关。

Abstract:Objective:To probe into the protective effect of total glucosides of paeony(TGP) on the mice with acute liver injury induced by carbon tetrachloride(CCl4).Methods:A total of 90 mice were randomly divided into the normal control group,the model control group,the high-dose TGP group,the middle-dose TGP group,the low-dose TGP group and dicyclohol tablet group according to their body weight.Each dose group of TGP was gavaged with 15 μL·g-1,while the normal control group and the model control group were given with equal volume of normal saline,and the dicyclohol tablet group with 110 mg·kg-1 dicyclohol tablet solution.Once a day,and for 7 days continuously.CCl4 induced chemical liver injury model was established,and 24 hours after the establishment of the model,0.4 mL of blood was collected from the orbital venous plexus of the mice.The activities of alanine aminotransferase(ALT),aspartate aminotransferase(AST) in the serum were detected by the kit method.The protein content of malondialdehyde(MDA),superoxide dismutase(SOD) and glutathione peroxidase(GPX) in the liver homogenate were detected by Lowry method.HE staining was used to make pathological examination and analysis of the liver tissue of mice.Results:The levels of ALT and AST in the model control group was higher than those in the normal control group,while the levels of ALT and AST in each dose group of TGP and the dicyclohol tablet group were lower than those in the model control group,and all the differences were statistically significant(P<0.05).The MDA level of liver homogenate in the model control group was higher than that in normal control group,while the contents of SOD and GPX were lower than those in normal control group,and all the differences were statistically significant(P<0.05).After administration of TGP with different dose,the increased level of MDA decreased to some degree.The MDA level of each TGP group with obviously increased liver homogenate was lower than that of model control group,and the contents of SOD and GPX were higher than those of the model control group,and all the differences were statistically significant(P<0.05).The MDA level of the dicyclohol tablet group was lower than that of model control group,and the difference was statistically significant(P<0.05).Conclusion:TGP has a definite protective effect on CCl4 induced acute liver injury in mice.Its mechanism is closely related to the regulation of immunity and antioxidation.
