藤黄酸及其衍生物抗肿瘤机制研究进展

摘    要:藤黄酸及其衍生物是存在于中药藤黄中的一类具有抗肿瘤活性的化合物。近年来,该类化合物药理学功效被国内外学者广泛研究,尤其抗肿瘤作用及相关机制已成为新近研究的热点。传统认为细胞毒是该类化合物抗肿瘤主要作用途径,但是目前的研究显示细胞凋亡、结合肿瘤细胞膜转铁蛋白调控细胞死亡、调节细胞周期、抑制肿瘤血管的生成、诱导细胞发生自噬在藤黄酸及其衍生物抗肿瘤中亦发挥着重要作用。就国内外相关文献及本实验室关于新藤黄酸的研究,对藤黄酸及其衍生物抗肿瘤的特性及抗肿瘤的作用机制作一综述,为藤黄酸及其衍生物的深入研究提供理论依据。

Abstract:Gambogic acid and its derivate,isolated from gamboge,are one type of compound possessing antitumor activity in Chinese herbs.Recently,pharmacological efficacy of such type of compound was widely studied by researchers,especially the antitumor effect and related mechanisms. Traditionally,cytoxicity was the principal pathway involved in antitumor effect of gambogic acid and its derivates. However,recent study indicated that apoptosis-inducing,mediating cell death by combining to TRF in tumor cell membrane,restricting cell cycle ,inhibiting tumor vessel and inducing autophagy also played key role in antitumor effect of such type of compound. Based on previous research and recent study conducted in our laboratory, here we conclude the antitumor effect of gambogic acid and its derivates and underlying mechanisms,which could provide theory base for the further study the activity of gambogic acid and derivates.
