摘    要:目的:建立测定当归芍药散中芍药苷、白芍苷、阿魏酸和洋川芎内酯I含量的RP-HPLC方法。方法:采用HPLC-PDA联用法,YMC C18色谱柱(4.6mm×150mm,5μm),以乙腈-水(0.1%v/v甲酸)为流动相,流速1.0mL.min-1,柱温23.5℃,检测波长200~400nm,数据处理选择最大吸收波长分别为232nm(芍药苷、白芍苷)、322nm(阿魏酸)、280nm(洋川芎内酯I)。结果:4种成分的线性范围分别为:芍药苷99.5~348μg.mL-1,白芍苷46.4~162μg.mL-1,阿魏酸1.03~30.8μg.mL-1,洋川芎内酯I 1.01~50.6μg.mL-1,相关系数R2均>0.999,方法精密度、回收率均符合要求。结论:该方法简便,结果准确可靠,重复性好,组方中其他组分均不干扰测定,适用于当归芍药散及单味药中芍药苷、白芍苷、阿魏酸和洋川芎内酯I的含量测定。

Abstract:Objective:To establish a RP-HPLC method for determination paeoniflorin,albiflorin,ferulic acid,and senkyunolide I in Dangguishaoyao San.Methods:HPLC-PDA determination was performed on a YMC C18 column(4.6mm×150mm,5μm).The mobile phase was consist with acetonitrile-H2O(0.1% v/v formic acid),and flow rate was 1.0mL·min-1.The detection wavelength was 200~400nm,and the peak area was chosen under their maximum absorption wavelength for 232nm(paeoniflorin,albiflorin),322nm(ferulic acid),and 280nm(senkyunolide I).Results:The linear ranged from 99.5 to 348μg·mL-1 for paeoniflorin(R2=0.999 2),46.4 to162μg·mL-1 for albiflorin(R2 = 0.999 4),1.03 to 30.8μg·mL-1 for ferulic acid(R2=0.999 8),and 1.01 to 50.6μg·mL-1 for senkyunolide I(R2 = 0.999 6).All of the recoveries were between 96.9%~103.4%.Conclusion:The method is simple,accurate,and reliable.The method can be applied to determination paeoniflorin,albiflorin,ferulic acid,senkyunolide I in Dangguishaoyao San and Radix paeoniae alba,Radix Angelica sinensis,Rhizoma Ligusticum chuanxiong.
