摘    要:目的:考察6种大孔树脂对葶苈子总强心苷的分离富集分离性能,确定最佳工艺条件。方法:以总强心苷吸附量,洗脱率,洗脱物总强心苷含量为指标,采用紫外-可见分光光度法进行含量测定。结果:NKA-9型树脂对葶苈子中总强心苷具有较好的分离富集分离能力,其最佳的工艺条件为:葶苈子提取物质量浓度4 g.mL-1(生药量),最大吸附量3.54 mg.g-1,装柱径高比1/3,吸附流速2 mL.m in-1,水洗脱量3倍柱体积,洗脱剂80%乙醇,洗脱量3倍柱体积,洗脱速度1 mL.m in-1。结论:NKA-9型树脂分离富集总强心苷的最佳工艺稳定高效,可推广应用于生产。

Abstract:Objective: To establish the separation and enrichment technology for total cardiac glycoside from Semen descuraniae,6 types of macroporous adsorption resin were investigated.Method: The adsorption capacity,elution ratio and purify of total cardiac glycoside were used as evaluating criteria.The UV-VIS spectrophotometric method was used in the determination of total cardiac glycoside content.Result: NKA-9 had better property of separation and purification and the optimal technological conditions were: the concentration of the sample of Semen Descuraniae,4 g·mL-1(amount to crude drug),The maximum adsorption capacity of total cardiac glycoside,3.54 mg·g-1,the optimal column diameter/height,1/3,the flow velocity of adsorption,2 mL·min-1,washing water,3 times of resin volume,the eluting reagent,80% ethanol(3 times of the resin volume),eluting velocity,1 mL·min-1.Conclusion: The technology of separating and purifying total cardiac glycoside of Semen Descuraniae with NKA-9 was stable and effective,and can be applied to manufacture.
