小柴胡汤在Caco-2细胞模型的吸收特性和转运机制研究

摘    要:目的:研究小柴胡汤在Caco-2细胞模型的吸收特性和转运机制.方法:利用MTT实验筛选小柴胡汤在Caco-2细胞中的安全浓度,用Caco-2细胞模型研究小柴胡汤的双向转运情况,采用高效液相色谱法进行含量测定,考察时间对小柴胡汤吸收的影响.结果:汉黄芩苷(wogonoside,WGS)和汉黄芩一素(wogonin,WGN)由AP侧到BL侧的Papp分别为(1.23±0.09)×10-6,(1.07±0.89)×10-5,由BL侧到AP侧的Papp分别为(2.12±0.19)×10-6,(7.12±1.02)×10-6,二者的转运率随着时间的增加而增加,WGS和WGN的PappAP→BL/PappBL→AP分别为0.58,1.49.黄芩苷、黄芩素和甘草酸不能透过Caco-2细胞单层.结论:WGS和WGN在Caco-2,细胞单层模型中的转运途径可能为被动转运.
