淫羊藿苷促进体外培养大鼠骨髓间 充质干细胞的成骨性分化

摘    要:目的:研究淫羊藿苷对大鼠体外培养骨髓间充质干细胞(rat bone marrow stromal cells,rBMSCs)成骨性分化的影响.方法:贴壁筛选法体外培养rBMSCs,采用1×10-5mol·L-1的淫羊藿苷进行药物干预,比较淫羊藿苷组和不加药的对照组之间碱性磷酸酶活性、碱性磷酸酶阳性克隆数(CFU-FALP)及钙化结节数量,提取总RNA,RT Real-time PCR检测bFGF,IGF-1,Osterix(OSX),Runx-2 mRNA的表达情况.结果:1×10-5mol·L-1淫羊藿苷可提高碱性磷酸酶(ALP)活性,增加钙化结节和CFU-FALP数量,促进与成骨性分化相关因子bFGF,IGF-1,OSX,Runx-2 mRNA的表达.结论:淫羊藿苷可显著促进rBMSCs的成骨性分化,提示淫羊藿苷具有开发为抗骨质疏松或促进骨折愈合新药的潜力.

Abstract:Objective : To investigate the effects of icariin on the osteogenic differentiation of rat bone marrow stromal cells(rBMSCs). Method : rBMSCs were cultured by adherence screening method. Icariin was supplemented into the culture at 1×10-5 mol·L-1. The osteogenic differentiation markers including alkaline phosphatase (ALP) activity, CFU-FALP and mineralized bone modulus were compared between the icariin-supplemented group and the control group. Total RNA was isolated and the gene expression of bFGF, IGF-1, Osterix(OSX) and Runx-2 was investigated by RT Real-time PCR. Result : Icariin significantly improved ALP activity, CFU-FALP amounts and mineralized modulus. It also can enhance the mRNA level of bFGF, IGF-1, Osterix and Runx-2. Conclusion : Icariin enhances the osteogenic differentiation of rBMSCs significantly, which suggested that icariin has the potentiality to be a new drug of anti-osteoporosis or fracture healing.
