龙骨马尾杉环阿屯醇合成酶(HcCAS1)基因的 克隆和生物信息学分析

摘    要:目的:对大伸筋草的原植物龙骨马尾杉Huperzia carinata环阿屯醇合成酶(cycloartenol synthase,CAS)基因HcCAS编码区进行克隆及序列分析。方法:根据本实验室已报道的龙骨马尾杉转录组高通量测序结果分析获得1条具有环氧角鲨烯环化酶保守结构域的CAS基因序列,采用RT-PCR技术,以龙骨马尾杉根、茎、叶混合样品总RNA为模板克隆得到龙骨马尾杉HcCAS基因的编码区序列,并对HcCAS蛋白进行理化性质、蛋白二级结构及三维结构预测分析。结果:序列分析表明,所克隆的HcCAS基因编码区开放阅读框(open reading frame,ORF)长为2 274 bp,编码757个氨基酸残基,命名为HcCAS1(GenBank登陆号JN790125)。结论:该研究在国内外首次获得龙骨马尾杉HcCAS基因的编码区序列,为进一步研究HcCAS蛋白在石杉科植物甾醇合成途径中的功能及酶活性位点的鉴定奠定基础。
