延胡索乙素对大鼠低O_2高CO_2肺动脉高压及FAK、ERK和Caspase 3表达的影响

摘    要:目的:研究延胡索乙素对慢性低O2高CO2性肺动脉高压的作用及粘着酶激酶FAK、ERK和凋亡蛋白酶Caspase 3表达的影响。方法:常压低O2高CO2舱制备低O2高CO2肺动脉高压大鼠模型,延胡索乙素ip 40mg/kg。右心导管法测肺动脉压;光镜下测定肺细小动脉管壁面积/管总面积(WA/TA)、肺细小动脉中膜厚度(PAMT);免疫组化法检测细胞外信号调节激酶(ERK2)、Ⅰ型胶原的表达量;原位杂交法检测FAKmRNA、Caspase3 mRNA的表达量。结果:延胡索乙素组mPAP显著低于低O2高CO2组;延胡索乙素组的WT/TA、PAMT也显著低于低O2高CO2组;延胡索乙素组肺细小动脉ERK2、I型胶原的相对含量、FAKm-RNA、Caspase3 mRNA的表达均显著低于低O2高CO2组。结论:延胡索乙素能明显抑制肺动脉高压的形成,延缓肺动脉血管的重构,抑制FAK表达、增强Caspase3的表达可能是其重要的机制之一。
