摘    要:目的:探讨黄芪甲苷对人外周血内皮祖细胞的保护作用。方法:用密度梯度离心法获取单个核细胞(MNCs),培养7天后,收集贴壁细胞,采用多波长激光共聚焦显微镜(laserscanningeonfoealmicroscope,LSCM)鉴定后将细胞随机分成:(l)对照组;(2)辛伐他汀对照组;(3)黄芪甲苷组。24h后采用流式细胞仪检测EPCs的增殖周期;MTT比色法、黏附能力实验检测内皮祖细胞(En-dothelialprogenitoreells,EPCs)的增殖能力、黏附能力。结果:黄芪甲苷在10-1mmol/L~10-10mmol/L的浓度范围中均有效,在黄芪甲苷10-9mmol/L浓度时,EPCs的增殖力最强,黄芪甲苷能明显增强EPCs的黏附、增殖能力。结论:黄芪甲苷能够增强EPCs的生物学功能,对EPCs有保护作用。

Abstract:Objective: To explore the effects and mechanisms of astragaloside mobilizing peripheral blood endothelial progenitor cells.Methods: Peripheral blood mononuclear cells(PB-MCs) were isolated by Ficoll density gradient centrifugation,and EPCs were identified by laserscanningeonfoealmicroscope(LSCM).Adherent cells were collected after seven days’ incubation and randomly divided into 3 groups(Control,simvastatin,astragaloside).The adhesion activity was determined by cell counts.The proliferation was determined by MTT assay and cell proliferation cycle analysis assay.Result: Compared with the control group,the numbers of EPCs adhesion,proliferation and cell proliferation cycle in astragaloside group and simvastatin group were significant(P < 0.01).The proliferative capacities of EPCs were improved in astragaloside group from 10-1mmol / L to 10-10mmol / L,significantly in 10-9mmol / L group.Conclusion: astragaloside has a significant protective effect of EPC.
