丹莪妇康煎膏对小鼠痛经模型的影响

摘    要:目的:观察丹莪妇康煎膏对缩宫素诱发小鼠痛经的镇痛作用。方法:小鼠随机分为空白对照组、模型组、少腹逐瘀丸组、丹莪妇康煎膏高剂量组及低剂量组。除空白对照组外,其余各组小鼠灌胃给药同时灌服己烯雌酚,连续7天,末次给药30分钟后,除空白对照组外,其余各组小鼠腹腔注射缩宫素,观察小鼠离体子宫平滑肌收缩变化、小鼠30分钟内扭体反应次数,并检测小鼠血浆血栓烷B2(TXB2)、6-酮-前列腺素F1α(6-Keto-PGF1α),血清超氧化物歧化酶(SOD)、丙二醛(MDA)及子宫组织一氧化氮(NO)、内皮素(ET)含量。结果:高剂量丹莪妇康煎膏(5.6g/kg体重)能显著抑制缩宫素诱导小鼠子宫平滑肌收缩,明显减少小鼠扭体次数,明显降低小鼠血浆TXB2、血清MDA及子宫组织ET值,明显升高血浆6-Keto-PGF1α及子宫组织NO水平,虽能使血清SOD值有升高的趋势,但无统计学意义;低剂量丹莪妇康煎膏(2.8g/kg体重)能明显抑制缩宫素诱导小鼠子宫平滑肌收缩张力,显著降低血浆TXB2水平,虽能使小鼠扭体次数、血清MDA和子宫组织ET值有下降的趋势,血浆6-Keto-PGF1α、血清SOD和子宫组织NO值有升高的趋势,但无统计学意义。结论:丹莪妇康煎膏对缩宫素诱导小鼠痛经模型有较好治疗作用,其作用可能与调节TXB2与6-Keto-PGF1α、NO与ET比值、抗氧化有关。

Abstract:Aim: To investigate the effect and possible mechanism of Dan’e Fukang decocted extrace(DFD) on oxytocin-induced dysmenorrhea in mice.Methods: Fifty mice were randomly divided into 5 groups,control group,dysmenorrhea model group,Shaofu Zhuyu Pill group,high dose DFD group and low dose DFD group.All groups except control group mice were administrated drug and diethylstilbestrol for 7 days.After intraperitoneal injection oxytocin in every groups except control group mice,the contractile amplitude of uterine smooth muscles and the frequency of strething writhing was recorded,and the levels of plasma TXB2,6-Keto-PGF1α,serum SOD and MDA,NO and ET in uterine tissue were determined.Results: High-dose DFD(5.6g/kg) could significantly inhibit the contractile amplitude of uterine smooth muscles(P<0.01),decreased the frequency of strething writhing(P<0.05),decreased the levels of plasma TXB2,serum MDA and ET in uterine tissue(P<0.05),and increased the level of plasma 6-Keto-PGF1α(P<0.05)and NO in uterine tissue(P<0.01),but there was no significant difference in the level of serum SOD compared with control group(P>0.05).Low-dose DFD(2.8g/kg) could inhibit the contractile amplitude of uterine smooth muscles(P<0.05),and significantly decreased the level of plasma TXB2(P<0.01),but there was no significant difference in the frequency of strething writhing and the levels of serum MDA,SOD,plasma 6-Keto-PGF1α,NO and ET in uterine tissue compared with control group(P>0.05).Conclusion: DFD has a better effect on dysmenorrhea mice induced by oxytocin through regulating TXB2/6-Keto-PGF1α,NO/ET ratio and antioxdant.
