山茱萸免疫活性部位F-1C的免疫抑制作用

摘    要:目的:研究山茱萸免疫活性部位F-1C对小鼠免疫反应的抑制作用。方法:运用淋巴细胞增殖反应、混合淋巴细胞反应、溶血空斑实验和流式细胞技术观察F-1C体外应用和口服对正常小鼠免疫反应和脾脏T细胞亚群的影响。结果:F-1C在10-200μg/ml剂量范围内无明显细胞毒性,但对小鼠ConA诱导的脾细胞增殖反应、混合淋巴细胞反应和脾细胞抗体生成反应具有明显的抑制作用,对LPS诱导的裸鼠脾细胞增殖反应呈剂量依赖性的促进作用。连续7天口服给予40和80mg/kg,明显抑制脾细胞抗体生成反应和脾细胞自发增殖反应,对脾脏T细胞亚群CD4^ 细胞数目无明显影响,但明显升高CD8^ 细胞百分数,使CD4^ /CD8^ 比值降低。同时给药组小鼠T抑制细胞(Ts)的抑制功能增强。结论:F-1C对正常小鼠免疫反应具有明显的抑制作用,增加Ts细胞数量并增强Ts细胞抑制功能可能是其发挥免疫抑制作用的途径之一。

Abstract:AIM:To investigate the immunosuppressive effect of F 1C,a fraction from the \%Cornus officinals\%, which is one component of Liu Wei Di Huang (LW) decoction. METHOD: The effects of F 1C on immune response were evaluated \%in vitro\% and \%in vivo\%. RESULT: F 1C showed strong inhibiting effect on antibody production response and proliferation of lymphocyte, during concentration range without cytotoxinic effect in vitro. Oral administration of F 1C also significantly inhibited antibody production response and splenocyte proliferation response. F 1C showed no influence on the number of CD 4 splenocyte, however, it increased the number of CD 8 splenocyte and decreased the ratio of CD 4 /CD 8 , suggesting that F 1C increased the quantity of Ts cell. The inhibition function of Ts was also enhanced by F 1C. CONCLUSION: F 1C possesses potent immunosuppressive effect. Enhancing the activity and quantity of Ts cell is one possible underlying mechanism.
