加减四逆散对慢性心理应激大鼠胸腺糖皮质激素受体作用的研究

摘    要:目的:观察加减四逆散对慢性心理应激大鼠胸腺细胞糖皮质激素受体作用的影响。方法:将大鼠随机分为正常对照组(C)、模型组(M)、加减四逆散组(C1)、人参皂甙组(C2)。电子天平称重并计算大鼠胸腺重量指数,采用放射免疫方法检测大鼠胸腺细胞糖皮质激素受体的数目、核转移率。结果:与C组比较,M组胸腺重量指数显著下降,糖皮质激素受体(glucocorticoid receptor,GCR)的数目略有上升的趋势,胸腺糖皮质激素受体的核转移率显著上升(P<0.01)。与M组相比,C1组与C2组的大鼠胸腺细胞糖皮质激素受体复合物的核转移率均显著降低,胸腺重量指数显著提高(P<0.01或P<0.05)。在受体核转移率下降方面,C1组优于C2组(P<0.05)。此外,C1组各项数据与C组比较无统计学差异。结论:加减四逆散能显著减轻糖皮质激素对胸腺的抑制作用。其作用途径可能是通过抑制慢性心理应激大鼠GCR由胸腺细胞胞浆向胞核转位来实现的。

Study on Effect of Jiajian Sini San on Glucocorticoid Receptor in Thymocyte of Chronic Psychological Stressed Rats

Abstract:Objective: To observe the effect of Jiajian Sini San (JJSNS) on glucocorticoid receptor (GCR) in thymocyte of chronic psychological stressed rats. Methods: Rats were divided randomly into 4 groups, the control group (C), the model group (M), the JJSNS treated group (C 1) and the ginsenoside treated group (C 2). Weight of thymus gland was weighed by electro balance. Number of GCR in thymocyte was detected with radioimmunoassay (RIA) and its nuclear translocation rate (GCR NTR) was calculated. Results: Compared with Group C, the thymus weight index in Group M was lowered significantly (P<0 05), and GCR in thymocyte showed a tendency to rise with the GCR NTR elevation significantly (P<0 01). Compared with Group M, the thymus weight index was higher and GCR NRT was lower in Group C 1 and C 2 (P<0 05 or P<0 01), and JJSNS showed more effective in reducing GCR NTR than ginsenoside. The above mentioned parameters in Group C 1 were different from those in Group C insignificantly. Conclusion: JJSNS could weaken the inhibitory effect of glucocorticoid on thymus by means of suppressing the translocation activity of GCR from cytoplasm to nucleus in chronic psychological stressed rat.
