补肾安胎方调节胚泡着床障碍小鼠雌、孕激素的实验研究

摘    要:目的观察补肾安胎方对着床障碍小鼠妊娠率和着床点数的影响.方法将妊娠小鼠随机分为3组,于妊娠第1天正常组和模型组灌胃自来水,中药组灌胃补肾安胎方;于妊娠第3、4天每天2次正常组皮下注射溶剂,模型组和中药组分别注射吲哚美辛(4.33 mg/kg)造成小鼠胚泡着床障碍的动物模型,妊娠第5、8天留取血清;采用放免法检测3组小鼠雌、孕激素含量;妊娠第8天观察比较3组小鼠妊娠率、妊娠小鼠着床点数;采用免疫组化技术检测子宫雌、孕激素受体表达水平.结果(1)小鼠妊娠率和胚泡着床点数:模型组(27.3%、5.3±0.7)显著低于正常组(90.9%、13.3±2.8,P<0.01),中药组(72.7%、10.7±2.2)较模型组显著增高(P<0.05).(2)血清雌激素水平:妊娠第5、8天时中药组均高于模型组(P<0.05或P<0.01),第8天时高于正常组(P<0.01).(3)血清孕激素水平:妊娠第5、8天时模型组、中药组均低于正常组(P<0.05或P<0.01),但第8天中药组显著高于模型组(P<0.01).(4)免疫组织化学结果:中药组雌、孕激素受体表达显著高于模型组.结论补肾安胎方能提高吲哚美辛诱导着床障碍小鼠妊娠率和着床点数,其机制可能与该方能改善甾体激素水平,促进子宫内膜雌、孕激素受体的表达有关.

Study on Regulatory Effect of Bushen Antai Recipe on Levels of Estrogen and Progesterone in Blastocyst Implantation Dysfunction Mice

Abstract:OBJECTIVE: To investigate the effect and explore the mechanism of Bushen Antai Recipe (BAR) on pregnancy rate and number of implantation site in blastocyst implantation dysfunction (BID) mice induced by indomethacin. METHODS: Pregnant mice were divided into 3 groups randomly: the normal group, the model group and the BAR group. Tap water was given orally to the rats in the normal and model groups, and BAR to the rats in the BAR group from the first day of pregnancy for 5 or 8 days; on the 3rd and 4th day dissolvent was injected subcutaneously twice per day in the normal group, while indomethacin (4.33 mg/kg) was injected subcutaneously twice per day in the other two groups to establish implantation dysfunction model; serum estrogen (E) and progesterone (P4) levels were detected on the 5th and 8th day; the pregnancy rate and number of implanted site was observed and the receptors of E and P4 in endometrium of uterus were examined by immunohistochemistry on the 8th day. RESULTS: The pregnancy rate and number of implanted site was 27.3% and 5.3 +/- 0.7 respectively in the model group, significantly lower than those in the normal group (90.9%, 13.3 +/- 2.8), and the BAR group (72.7%, 10.7 +/- 2.2, P < 0.05). Serum E level was higher in the BAR group than that in the model group on the 5th and 8th day, and even higher than that in the normal group on the 8th day; serum P4 level was lower in the model and BAR groups than that in the normal group on the 5th day (P < 0.01), but higher in the BAR group than that in the model group on the 8th day. Immunohistochemical observation showed that expressions of E and P4 receptor increased remarkably in the BAR group than those in the model group. CONCLUSION: BAR increases the pregnancy rate and number of implanted site of indomethacrne induced BID mice through regulating E and P4 levels and enhancing the expressions of their receptors in the endometrium.
