重肌灵片对实验性自身免疫性重症肌无力大鼠的影响

目的观察重肌灵片对实验性自身免疫性重症肌无力(EAMG)的影响。方法用从电鳐电器官提取的含 N_2-AchR 粗提液加完全弗氏佐剂免疫 Lewis 大鼠制成 EAMG,动物于第2次免疫后第5天开始给药,对 EAMG 大鼠的临床症状、肌力、运动能力、肌电图,血清中抗 N_2-AchR-Ab、突触后膜 N_2-AchR 的数目以及淋巴细胞转化等指标进行观察。结果重肌灵片可以改善 EAMG 大鼠的临床症状,提高该鼠体重、肌力和运动能力,抑制血清中抗 N_2-AchR-Ab,降低连续低频电刺激后出现的肌肉收缩幅度的衰减,提高突触后膜 N_2AchR 数目。抑制 N_2-AchR 诱导的 EAMG 大鼠特异性淋巴细胞增殖反应,但对 ConA 诱导的非特异性增殖反应有一定增强作用(P<0.01或P<0.05)。结论重肌灵片药理作用不同于西药的免疫抑制剂,具有选择性免疫调节作用,即下调针对 N_2-AchR 特异性病理免疫性反应,对正常免疫功能不抑制反而增强。

Effect of Zhongjiling Tablet in Treating Rats with Experimental Autoimmune Myasthenia Gravis

Objective To observe the effect of Zhongjiling tablet(ZJLT)in treating rats with experimental autoimmune myas- thenia gravis(EAMG).Methods EAMG was made by adding complete Freund's adjuvant in coarse extraction contained N2-AchR from Torpedo's electrical organ.Medication was started on the 5th day from the second immunization.Indexes as clinical symptoms,myodynamia,movement capability,electromyogram(EMG),serum anti-N_2-AchR-Ab,number of postsynaptic membrane,lymphocyte transformation,etc.were observed.Results ZJLT could improve the clinical symp- toms of EAMG rats,improve the rats'weight,myodynamia,movement capability,inhibit serum anti-N_2-AchR-Ab,lower the declination of muscular contraction amplitude after the stimulation by the continuous low-frequency electricity,in- crease the number of postsynaptic membrane,inhibit the specific lymphocyte transformation of EAMG rats induced by N_2- AchR.But it had certain enhancing effect on the nonspecific proliferation reaction induced by ConA(P<0.01 or P <0.05).Conclusion The pharmacological function of ZJLT was different from the immune inhibitor agents of western medicine.It has selective immune regulatory function,ie.down-regulating N_2-AchR specific pathological immune reac- tion while enhancing the normal immunity.
