实验性骨关节炎MMP-3与软骨细胞凋亡

目的:为观察实验性骨关节炎中MMP-3和软骨细胞凋亡情况,并对二者表达作相关分析。方法:采用改良Hulth’s法制造兔骨关节炎模型,分别在8周、16周抽取关节液测定MMP-3的含量,切取关节软骨。采用TUNEL法标记凋亡软骨细胞计数,并对软骨进行HE染色和透视电镜观察。结果:HE染色中,正常组软骨细胞排列规则,关节炎组中软骨细胞排列紊乱。透视电镜观察各组均可见凋亡细胞,但在关节炎组中可见胶原纤维失去正常的结构,排列紊乱,可见钙盐沉积。在MMP-3测定中,早期骨关节炎(20.91±2.48)ng.mL-1和晚期骨关节炎(41.68±5.71)ng.mL-1均高于正常对照组(4.34±2.13)ng·mL-1,差异有统计学意义P0.01;晚期骨关节炎和早期关节炎组比较(P0.01);早期骨关节炎(10.18±1.90)和晚期骨关节炎(35.47±3.46)软骨细胞凋亡指数都高于正常对照组(3.93±1.91),P0.01;晚期骨关节炎早期关节炎组比较,(P0.01)。MMP-3变化和软骨细胞的凋亡在各个时期均存在相关性(r=0.82,P=0.03;r=0.64,P=0.049;r=0.69,P0.029)。结论:实验性骨关节炎中,关节滑液中MMP-3含量在模型组中明显升高,晚期含量升高更明显;软骨细胞的凋亡指数和MMP-3的变化密切相关,MMP-3的升高可能是导致软骨基质破坏,软骨细胞凋亡的重要因素。

To observe the MMP-3 in joint synovial fluid and chondrocyte's apoptosis of experimental osteoarthritis

Objective：To observe the correlation between MMP-3 of joint synovial fluid and chondrocyte’s apoptosis in experimental osteoarthritis.Methods：OA model was induced in rabbits by Hulth’s method. The MMP-3 of knee Joint synovial fluid were examined at the end of 8 and 16 weeks. The changes of morphology of chondrocyte and cartilage were observed by light microscopy and transmission electron microscopy and apoptosis examined by TUNEL.Results：Compared with early model stage group （20.91±2.48）ng·m^-1, MMP-3 expression was significantly increasing in late model stage group （41.68±5.71） ng·m^-1, P〈0.01. Apoptosis cell index had significant difference between early model stage 10.18±1.90 and late model stage 35.47±3.46,P〈0.01. Apoptosis cell index and MMP-3 has significant correlation between normal groups and model group at every stage.Conclusion：MMP-3 increased in knee joint fluid of experimental osteoathritis, especially in late stage. MMP-3 may be one of the possible reason of ECM destruction and apoptosis of chondrocytes.
