通痹灵胶囊对体外培养的强直性脊柱炎成纤维细胞的影响

为观察中药通痹灵胶囊及含药血清对体外培养的强直性脊柱炎 (AS)成纤维细胞增殖的影响 ,探讨其药理作用机制 ,采用体外细胞培养技术 ,以AS患者髋关节囊成纤维细胞为材料 ,采用四甲基偶氮唑盐 (MTT)检测法、3 H TdR掺入法、流式细胞仪、绘制成纤维细胞生长曲线等方法观察中药通痹灵胶囊对AS成纤维细胞增殖、DNA合成及生长周期的影响。结果显示通痹灵胶囊浓缩液及含药血清不仅对成纤维细胞的增殖有明显抑制作用 (P <0 .0 1) ,还对其DNA的合成有抑制作用 (P <0 .0 1) ,流式细胞仪检测显示中药组G0 G1 期细胞增多 ,S期细胞明显减少 (P <0 .0 1)。说明中药通痹灵胶囊及其含药血清对体外培养AS成纤维细胞增殖所产生的明显抑制作用 ,可能是其治疗AS的药理机制之一。

THE EFFECT OF TONGBILING CAPSULE INHIBITING THE PROLIFERATION IN CULTURED AS HIP JOINT BURSA FIBROBLAST

In order to study the effect of TongBiling capsule inhibiting the proliferation in Ankylosing Spondylitis(AS)hip joint bursa fibroblast cultured in vitro. Using techniques of cell culture in Vitro, we used the cultured AS hip joint bursa fibroblast as an experimental model. The cell growth curve, MTT assay, 3 H TDR endosmosis method and flow cytometry methed were used in this study to detect effect of the concentrated TongBi and the serum with TongBi on the ASfibroblast proliferation, DNA synthesis, cell cycle. The results thowed that both the concentrated TongBi and the serum with TongBi could ont only significantly inhibit the proliferation, but also reduce the DNA synthesis ofthe cultured AS fibroblast(P<0.01).The cell cycle determinated by FCM analysis showed that the cell proportion G 0 G 1 phase increased and in S phase decreased( P <0.01).It suggested that TongBi could significantly inhibit the proliferationand DNA synthesis in cultured AS fibroblasts. This may be one of pharmacologic mechanism of TongBi capsule in the treatment of AS.
