电针对干眼症兔角膜TNF-α、IL-6、MHC-Ⅱ调节作用的研究

摘    要:目的通过观察电针对干眼症兔角膜组织肿瘤坏死因子-α(TNF-α)、白细胞介素-6(IL-6)和组织相容性复合体-Ⅱ(MHC-Ⅱ)表达的影响,探讨电针治疗干眼症的作用机制。方法24只雄性新西兰兔随机分为正常组、模型组、电针组和西药组,每组6只。正常组不造模、不干预。模型组仅给予0.1%苯扎氯铵滴眼造模,造模成功后不予任何干预处理。电针组造模后给予电针干预,取穴为双侧攒竹、太阳、丝竹空和风池,攒竹和太阳连接电针。西药组造模后给予聚乙二醇滴眼液治疗。实验结束后检测泪液分泌量和泪膜破裂时间,应用免疫组化法检测角膜组织TNF-α、IL-6和MHC-Ⅱ蛋白的表达。结果与正常组比较,模型组兔泪液分泌量降低,泪膜破裂时间缩短(P<0.01);与模型组比较,电针组兔泪液分泌量增多、泪膜破裂时间延长(P<0.01),西药组泪液分泌量无明显变化(P>0.05)、泪膜破裂时间延长(P<0.01)。与正常组比较,模型组兔角膜组织TNF-α、IL-6和MHC-Ⅱ蛋白表达均增加(P<0.05);与模型组比较,电针组、西药组兔角膜组织TNF-α、IL-6和MHC-Ⅱ蛋白表达均减少(P<0.05)。结论电针可以改善实验性干眼症兔泪液分泌量和泪膜破裂时间,下调角膜组织TNF-α、IL-6和MHC-Ⅱ的表达。

Abstract:Objective By observing the effects of electroacupuncture(EA)on the expressions of tumor necrosisfactor(TNF)-α,interleukin(IL)-6 and major histocompatibility complex(MHC)-Ⅱin the cornea tissues of rabbits withdry eye syndrome(DES),to discuss the mechanism of EA in the treatment of DES.Method Twenty-four male NewZealand rabbits were randomly divided into a normal group,a model group,an EA group and a Western medicationgroup,with 6 rabbits in each group.The normal group did not undergo modeling process or any intervention.Themodel group only received eye drop of 0.1%benzalkonium chloride to develop models,but did not undergo anyintervention afterwards.The EA group was given EA intervention after modeling by selecting bilateral Cuanzhu(BL2),Taiyang(EX-HN5),Sizhukong(TE23)and Fengchi(GB20)with Cuanzhu(BL2)and Taiyang(EX-HN5)connecting toEA apparatus.The Western medication group was intervened by Polyethylene glycol eye drops after modeling.After the end of experiment,the tear production and tear film break-up time(BUT)were measured.Immunohistochemistry method was used to detect the expressions of TNF-a,IL-6 and MHC-Ⅱproteins in cornea tissues.Result Compared with the normal group,the rabbit tear production decreased and the BUT was shortened in the model group(P<0.01).Compared with the model group,the rabbit tear production increased and the BUT was prolonged in the EA group(P<0.01);compared with the model group,the Western medication group did not show significant change in tear production(P>0.05)but showed prolonged BUT(P<0.01).Compared with the normal group,the expressions of TNF-a,IL-6 and MHC-Ⅱproteins in cornea tissues increased in the model group(P<0.05).Compared with the model group,the expressions of TNF-a,IL-6 and MHC-Ⅱproteins in cornea tissues decreased in the EA group and the Western medication group(P<0.05).Conclusion EA can improve tear production and BUT in experimental DES rabbits,and down-regulate the expressions of TNF-a,IL-6 and MHC-Ⅱ proteins in cornea tissues.
