针刺捻转补泻手法对自发性高血压大鼠小脑GABA、Glu表达的影响

摘    要:目的通过观察针刺捻转补泻手法对自发性高血压大鼠(SHR)小脑组织中γ-氨基丁酸(GABA)、谷氨酸(Glu)含量表达的影响,探讨其对SHR大鼠小脑的保护机制,为捻转补泻降压效应差异提供客观依据。方法雄性SHR大鼠24只随机分为捻转补法组、捻转泻法组、电针组、模型组,每组6只。6只Wistar-Kyoto(WKY)大鼠作为空白组。空白组和模型组每天进行与各针刺组相同的抓捉固定刺激,其余各针刺组分别采用相应治疗手法针刺双侧“太冲穴”,每日上午治疗1次。用无创血压仪测量各组大鼠尾动脉收缩压,第1、4、7、11、14日测量并记录。针刺处理14 d后,分别取大鼠小脑组织,采用酶联免疫吸附法(ELISA)分别检测小脑组织中GABA、Glu的含量并进行统计学分析。结果1)模型组大鼠收缩压与空白组相比显著升高(P<0.01);与模型组比较,3个针刺组均能明显降低SHR大鼠血压(P<0.01),且捻转泻法组血压降低最显著,与模型组及其他治疗组相比差异显著(P<0.01);捻转补法组与电针组比较无统计学意义(P>0.05)。2)与空白组比较,模型组小脑组织中GABA含量明显降低(P<0.01),Glu含量明显升高(P<0.01);捻转泻法组小脑组织中GABA含量较模型组、捻转补法组和电针组明显升高(P<0.05或P<0.01),Glu含量较模型组、捻转补法组和电针组明显降低(P<0.05或P<0.01);捻转补法组与电针组比较差异无统计学意义(P>0.05)。结论SHR大鼠小脑组织中GABA含量的降低与Glu含量的升高可能是高血压形成的中枢机制之一,对“太冲穴”施以针刺捻转补泻手法可有效降低血压,使GABA含量升高、Glu含量降低,小脑中GABA、Glu参与了针刺捻转补泻手法对SHR的血压调节过程,且不同针刺手法调节作用差异显著,捻转泻法效果最显著。

Abstract:Objective:To explore the mechanism of cerebellum protection in spontaneously hypertensive rats(SHR),and to provide objective basis for the difference of hypotensive effect of the twisting for reinforcing and reducing methods in acupuncture by observing its effect on GABA and Glu in cerebellar tissue of SHR.Methods:24 male SHR were randomly divided into two groups:the reinforcing group,the reducing group,electroacupuncture group and the model group,6 rats in each,and six Wistar-Kyoto(WKY)rats were used as the blank group.The blank group and the model group were given the same grasping and fixation stimulation as the acupuncture groups every day.The acupuncture groups received acupuncture treatment on bilateral Taichong point(LV3)once a day in the morning with corresponding manipulation methods.The systolic blood pressure of the tail artery in each group was measured by non-invasive blood pressure meter on the 1st,4th,7th,11th and 14th day after acupuncture.After 14 days of acupuncture treatment,the cerebellum tissues of rats were taken and the contents of GABA and Glu in cerebellum were detected by ELISA and analyzed statistically.Results:1)The systolic blood pressure of the model group was significantly higher than that of the blank group(P<0.01);compared with the model group,the blood pressure of SHR rats in the three acupuncture groups were significantly reduced(P<0.01),and the blood pressure dropped the most in the reducing group,which was significantly different from the model group and the other treatment groups(P<0.01);there was no statistical difference between the reinforcing group and the electroacupuncture group(P>0.05).2)Compared with the blank group,the GABA content in the cerebellar tissue of the model group was significantly reduced(P<0.01),and the Glu content increased significantly(P<0.01),with the difference statistically significant;the GABA content in cerebellar tissue of the reducing group was significantly higher than that of the model group,the reinforcing group and the electroacupuncture group(P<0.01 or P<0.05);the Glu content was significantly lower than that of the model group,the reinforcing group and the electroacupuncture group(P<0.01 or P<0.05);there was no statistical difference between the reinforcing group and the electroacupuncture group(P>0.05).Conclusion:The decrease of GABA and the increase of Glu in cerebellar tissue of SHR rats may be one of the central mechanisms of hypertension formation.The application of twisting for reinforcing and reducing methods in acupuncture on the Taichong point(LV 3)can lower blood pressure,increase GABA and decrease Glu effectively.GABA and Glu in the cerebellum have participated in the regulation process of twisting for reinforcing and reducing methods in acupuncture to the blood pressure of SHR rats.The regulation effects vary significantly in different acupuncture manipulation methods,and the effect in twisting-for-reducing method group is the most significant.
