黄芪甲苷对射血分数保留型心力衰竭大鼠心脏功能保护及心肌微血管炎症因子的影响

摘    要:目的观察黄芪甲苷对射血分数保留型心力衰竭(HF)模型大鼠的心脏功能保护及其对心脏微血管和炎症因子的影响。方法将25只SPF级雄性盐敏感(Dahl-ss)大鼠随机分为5组:正常对照组、模型组及黄芪甲苷低、中、高剂量(10、20、40 mg/kg)组。观察各实验组心电图、心脏B超、血清NT-ProBNP、VEGF、CD31、vWF、TNF-α和IL-6的定位及表达。结果与模型组相比,黄芪甲苷低、中、高剂量组大鼠LVEF均值显著升高(P<0.05),且黄芪甲苷高剂量组效果最佳;血清NT-ProBNP降低(P<0.05),且剂量越大,降低越明显;VEGF、CD31表达明显增加(P<0.05);vWF、TNF-α和IL-6蛋白表达下调(P<0.05)。结论黄芪甲苷对HFpEF模型大鼠的心脏功能具有保护作用,其机制可能与抑制心肌组织炎性反应、改善心肌微血管功能和心肌损伤有关。

Abstract:Objective:To observe the protective effect of AstragalosideⅣon cardiac function in rats with heart failure with preserved ejection fraction(HFpEF)and its effects on cardiac micro vessels and inflammatory factors.Methods:25 SPF salt-sensitive male(Dahl-SS)rats were randomly divided into 5 groups:normal control group,model group and AstragalosideⅣlow-dose,medium-dose and high-dose(10,20 and 40 mg/kg)groups.Some indicators were observed including the location and expression of electrocardiogram,cardiac B-ultrasound,serum NT-ProBNP,VEGF,CD31,vWF,TNF-αand IL-6 in each experimental group.Results:Compared with the model group,the mean LVEF of rats in the AstragalosideⅣlow,medium,and high dose groups was significantly increased(P<0.05),and the AstragalosideⅣhigh dose group had the best effect.Serum NT-ProBNP decreased(P<0.05),and the higher the dose,the larger the decrease,and the obvious increase in VEGF and CD31 expression(P<0.05);the down-regulation of v WF,TNF-αand IL-6 protein expression(P<0.05).Conclusion:AstragalosideⅣhas a protective effect on the cardiac function of HFpEF model rats,and its mechanism may be related to inhibiting the inflammatory response of myocardial tissue,improving myocardial microvascular function and myocardial injury.
