复方薤白胶囊对不同时期肺动脉高压大鼠肺血管内皮细胞保护机制的探讨

摘    要:目的 探讨复方薤白胶囊对不同时期肺动脉高压大鼠肺血管内皮细胞的保护机制.方法 (1)8只SD大鼠予以随机编号后采用腹腔注射野百碱的方法制作肺动脉高压动物模型,每周处死2只大鼠检测肺小动脉内皮细胞的凋亡,共4周.(2)24只SD大鼠随机分为4组:正常对照组、模型组、复方薤白胶囊高剂量组、复方薤白胶囊低剂量组.采用腹腔注射野百碱的方法制作肺动脉高压动物模型,于造模第2日开始给予复方薤白胶囊灌胃干预1~4周,根据之前肺动脉高压大鼠模型凋亡的变化趋势选择凋亡达峰周和凋亡抑制明显周,各组处死3只大鼠进行肺血管内皮细胞凋亡的检测;第4周测定各组大鼠平均肺动脉压和平均右心室压.结果 (1)模型组大鼠第1周肺血管内皮细胞Caspase3表达达峰值,之后Caspase3的表达逐渐减少而血管内膜增生逐渐增加,各周时间点差异有统计学意义(P<0.05).(2)模型组的mPAP和mRVP明显升高,与正常对照组差异有统计学意义(P<0.01),复方薤白胶囊高剂量组和低剂量组mPAP和mRVP较正常组高但较模型组则显著下降(P<0.05).(3)与正常组相比,模型组大鼠肺血管内皮细胞Caspase3的表达在第1周明显升高,复方薤白胶囊高、低剂量组较模型组则明显减少(P<0.05);模型组大鼠肺血管内皮细胞Caspase3的表达在第4周较第1周明显减少,复方薤白胶囊高、低剂量组凋亡较模型组则明显增多(P<0.05).结论 复方薤白胶囊能有效降低MCT诱导肺动脉高压大鼠的肺动脉压,这可能与其在肺动脉高压形成早期抗内皮细胞凋亡和肺动脉高压晚期促进内皮细胞凋亡有关.

The Investigation of the Protection Mechanism of Compound Macro-stem Onion Capsule on Pulmonary Endothelial Cell in Different Periods of Rats with Pulmonary Hypertension

Abstract:Object:To investigate the effect of the Compound Macro-stem Onion Capsule on the apoptosis of pulmonary endothelial cell in different period of rats with pulmonary hypertension.Methods:(1) 8 SD rats were made into animal models of pulmonary hypertension by intra-peritoneal injection of wild lily alkali method,and 2 rats were executed for detection of small pulmonary artery endothelial cell apoptosis every week; (2)24 SD rats were randomly divided into four groups:①the normal control group;②the model group;③the high dose of Compound Macro-stem Onion Capsule group;④the low dose of Compound Macro-stem Onion group.Animal model of pulmonary hypertension was made with intra-peritoneal injection of wild lily alkali (MCT).Two days later,drug was given with intra-gastric injection.According to the results above,3 rats of each group were executed for the detection of small pulmonary artery endothelial cell apoptosis when the data arrived at highest or lowest level.Average pulmonary arterial pressure (mPAP) and average right ventricular pressure and average (mRVP) were determined after four weeks.Results:(1) The expression of Caspase3 in pulmonary vascular endothelial cell of model group rats reached the peak at the first week.After that,the expression of Caspase3 decreased and vascular intimal hyperplasia increased gradually.There was significant difference between the detection results of each week (P < 0.05).(2)Compared with the normal control group,the mPAP and mRVP of model group were obviously higher and the difference was significant(P< 0.01) ;the mPAP and mRVP of high dose and low dose of Compound Macro-stem Onion Capsule groups were significantly higher than that of the normal control group and lower than that of the model group (P < 0.05).(3) Compared with the normal control group,the expression of Caspase3 of the model group increased significantly in the first week,which was significantly higher than that of the high dose and low dose of Compound
