解毒化瘀Ⅱ方对实验性肝衰竭大鼠肝脏坏死面积及肝线粒体超微结构的影响

摘    要:目的:研究解毒化瘀Ⅱ方对实验性暴发性肝衰竭大鼠肝脏坏死面积及肝线粒体超微结构的影响。方法:将84只Wistar大鼠随机分为正常组,模型组,解毒化瘀Ⅱ方低、中、高剂量组,中药安宫牛黄丸组,西药乳果糖组;除正常组外各组均用皮下注射硫代乙酰胺(TAA)造成大鼠实验性暴发性肝衰竭模型,观察各组大鼠造模后12h肝脏的大体结构、病理组织学改变、坏死面积及肝线粒体超微结构。结果:模型组肝细胞大量坏死,肝线粒体高度水肿、坏死;解毒化瘀Ⅱ方各组能改善肝细胞、肝线粒体的病理损伤程度,减少坏死面积,与模型组比较有显著性差异(P<0.05或P<0.01),并呈现出量效关系;解毒化瘀Ⅱ方高剂量组在减少坏死面积、改善肝线粒体超微结构作用上优于乳果糖组、安宫牛黄丸组(P<0.05或P<0.01)。结论:解毒化瘀Ⅱ方对肝衰竭大鼠肝细胞、线粒体具有较好的保护作用。

Abstract:Objective: To study the effects of JieDuHuaYu II Decoction on liver necrotic area and mitochondrion ultrastructure in rats with experimental hepatic failure. Methods: 84 Wistar rats were randomly divided into the normal group, the control group, the JieDuHuaYu II groups of low, middle and high doses, the AnGongNiuHuang group, and the lactulose group. Except the normal group, the other groups were made into the models with experimental hepatic failure by injecting TAA. 12 hours later, the observation was made to see the change of structure and histopathology, the necrotic area, and the mitochondrion ultrastructure in the rats' livers. Results: In the control group, most of liver cells died, and the severe edema or even apoptosis were found in mitochondria; in the JieDuHuaYu II groups, the pathological injuries of liver cells and mitochondria were improved and the necrotic area was reduced with obvious differences (P<0.05 or P<0.01), and there was relationship of quantity-effect; better effects on reducing the necrotic area and improving the mitochondrion ultrastructure in the JieDuHuaYu II group of high dose than in the AnGongNiuHuang group and the lactulose group (P<0.05 or P<0.01). Conclusions: Better protective effects on liver cells and mitochondria can be achieved by JieDuHuaYu II Decoction.
