癌痛消胶囊调节小鼠荷H_(22)移植性肝癌细胞VEGF,p53和p21ras表达的实验研究

摘    要:目的:通过观察癌痛消胶囊调节小鼠荷H22 移植性肝癌细胞中血管内皮生长因子(Vascularen dothelialgrowthfactor,VEGF)、p53和p21ras的表达作用,以探讨该药抑制肿瘤的作用机理。方法:昆明种雄性小白鼠60只,按随机区组分为6组(A -F组) ;A -E组每鼠均于前肢右腋皮下接种0 2mlH22 肿瘤细胞悬液(浓度为1×107/ml) ,接种24h后按体重给药。A组、B组、C组分别给予大、中、小剂量的癌痛消胶囊,D组予康赛迪胶囊,E组予生理盐水,均灌胃给药,每日1次。连续用药10天,停药24h ,称重并处死动物,剥取瘤块称重,计算出肿瘤生长抑制率,用免疫组化法检测癌细胞中VEGF ,p53 ,p21ras ,VHL的表达程度。F组予饲料及水分正常饲养。结果:A ,B ,C ,D组均有明显的抑瘤作用,其中以A组的抑瘤作用最明显(P<0 05或P<0 01)。造模各组动物的肝癌细胞中均见VEGF ,p53和p21ras的阳性染色,但阳性表达程度不同,与B ,C ,E组比较,癌痛消胶囊大剂量组和康赛迪胶囊组均可明显降低肝癌细胞中VEGF ,p53和p21ras的阳性表达(P<0 05或P<0 01)。结论:癌痛消胶囊对小鼠H22 移植性肝癌有抑瘤作用,抑制肿瘤生长的机理可能与下调VEGF ,p53和p21ras的水平有关。

Abstract:Purpose:To observe the effects of Aitongxiao Capsule(ATX)on regulating the expression of vascular endothelial growth factor(VEGF),p53and p21ras in the transplanted hepatocarcinoma22(H 22 )tumor of mice,and explore its tuˉmor-inhibiting mechanism.Methods:60Kunming male mice were randomly divided into6groups(A,B,C,D,E,F).Then0.2ml H 22 suspension(1×10 7 /ml)was injected to each mouse in the first to five groups by right axillary subcutaneous vaccination in the forefoot.After24hours,the mice were treated with medicine according to their weight.Groups A,B and C were given high,medium and low dosages of ATX respectively,while Group D Kangsaidi Capsule(KSD)and Group E normal saline.All the medicine was given through gavage,1time/day.The treatment course lasted for10days.Then no medicine was given24hours,and immediately after that,the mice were weighted and killed to get their transplanted tumors to weigh and calculate the inhibitory rate of the tumor.Meanwhile,the expression of VEGF,p53and p21ras was determined by immunohistochemical method.During the whole process,the Group F was raised by normal forage and water.Results:The obvious inhibition of tumor was indicated in Groups A,B,C and D,with the Group A having the most obvious effect.(P<0.05or P<0.01).The positive staining of VEGF,p53and p21ras was found in all the treatment groups with different degree.When compared with Group B,C and E,the positive expression of VEGF,p53and p21ras was lowered in the high dosage ATX group and the KSD group distinctively(P<0.05or P<0.01).Conclusions:The ATX capsule has the inˉhibitory action for transplanted hepatocarcinoma22(H 22 )tumor of mice,and its mechanism for inhibiting the tumor growth may have correlation with the decrease of VEGF,p53and p21ras.
